  1. 1. Micro-scale Platforms for Investigations of Biological Structures

    Author : Anna Kim; Chalmers tekniska högskola; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; NATURVETENSKAP; NATURAL SCIENCES; microfabrication; non-ionic surfactants; single-cell viability; hydrodynamic flow confinement; cell-to-cell connections; SU-8; Teflon AF; cell protrusions; free-standing probes; millimeter-waves;

    Abstract : This thesis describes the development of innovative micro-scale platforms as a means to address the current challenges within the field of cell biology, and to provide an interface between miniaturized analytical technology and experimental systems for the life sciences. The work includes an analytical platform for rapid assessment of the viability of adherent single cells; a surface patterning strategy for investigations of cell-to-cell communication via protrusions; a versatile microfluidic probe in SU-8 hard-polymer, and a microfabricated millimeter-wave measurement system for the identification and characterization of niosome (non-ionic surfactant vesicle) constituents, which is operated in a label-free, non-invasive manner. 2. 2. In vitro and in silico Predictions of Hepatic Transporter-Mediated Drug Clearance and Drug-Drug Interactions in vivo

    Author : Anna Vildhede; Per Artursson; Maria Karlgren; Maria Kjellsson; Ulf Norinder; Kim Brouwer; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; OATP1B1; OATP1B3; OATP2B1; NTCP; drug transporters; human hepatocytes; atorvastatin; pitavastatin; proteomics; sandwich-cultured human hepatocytes; SCHH; mechanistic modeling; in vitro-in vivo extrapolation; transport inhibition; hepatic uptake; hepatocyte isolation; transporter contribution; Biopharmaceutics; Biofarmaci;

    Abstract : The liver is the major detoxifying organ, clearing the blood from drugs and other xenobiotics. The extent of hepatic clearance (CL) determines drug exposure and hence, the efficacy and toxicity associated with exposure. Drug-drug interactions (DDIs) that alter the hepatic CL may cause more or less severe outcomes, such as adverse drug reactions. READ MORE
