  1. 1. Leukemia chemotherapy : experimental studies on pharmacological optimisation

    Author : Michèle Masquelier; Karolinska Institutet; Karolinska Institutet; []
    Keywords : ;

    Abstract : Our main goal has been to identify new means to improve chemotherapy of acute leukaemia: 1. By using low-density lipoprotein (LDL) as a drug carrier to increase the selectivity of antileukemic drugs, based on high LDL uptake in acute myeloid leukemia (AML) cells. READ MORE

  2. 2. Pharmacology and resistance mechanisms of nucleoside analogues and topoisomerase II interactive agents : studies on human leukemia cells with a focus on cross-resistance

    Author : Kourosh Lotfi; Rolf Larsson; Linköpings universitet; []
    Keywords : Nucleoside analogue; cross-resistance; multidrug resistance; deoxycytidine kinase; leukemia; anthracyclines; P-glycoprotein; MEDICINE; MEDICIN;

    Abstract : The purpose of this thesis was to elucidate mechanisms of action and resistance of clinically relevant nucleoside analogues and topoisomerase interactive agents in human leukemia cell lines and leukernia cells isolated from peripheral blood of leukemia patients. Interactions and cross-resistance patterns of these different cytotoxic drug families were also studied since these drugs are usually administrated in combination in the clinic. READ MORE

  3. 3. Deoxyribonucleoside kinases in nuclear and mitochondrial DNA precursor synthesis : phosphorylation of anti-cancer nucleoside analogs in different subcellular compartments

    Author : Chaoyong Zhu; Karolinska Institutet; Karolinska Institutet; []
    Keywords : deoxyribonucleoside kinase; deoxyguanosine kinase; deoxycytidine kinase; nucleoside analogs; mitochondria; phosphorylation;

    Abstract : Nucleoside analogs have been developed as anticancer and antiviral agents. The main mechanism of action of nucleoside analogs is the incorporation of the corresponding analog triphosphate into DNA where it interferes with DNA replication. READ MORE

  4. 4. Identification and characterization of nucleoside and nucleotide kinases : pharmacological activation of anti-cancer and anti-viral nucleoside analogs

    Author : An R Van Rompay; Karolinska Institutet; Karolinska Institutet; []
    Keywords : ;

    Abstract : Cells require deoxyribonucleotides to synthesize DNA. Structurally modified precursors of deoxyribonucleotides, i.e. nucleoside analogs, are used as anti-viral and anti-cancer compounds. READ MORE

  5. 5. Exploring novel roles of metabolic enzymes MTHFD2 and PFKFB3 in cancer genome stability and their potential as anticancer therapeutic targets

    Author : Nadilly Y Bonagas Villarreal; Karolinska Institutet; Karolinska Institutet; []
    Keywords : ;

    Abstract : Altered tumor metabolism has been described as early as the 1920s, but it was only in recent decades that proteomic and metabolomic studies revealed that the ways in which tumors rewire their nutrient and energy pathways are more diverse and have more implications for treatment outcome than previously thought. There is now a great interest in characterizing promising metabolic targets and identifying novel ways by which to exploit them for cancer treatment. READ MORE
