  1. 1. Pharmacogenetic Studies of Paclitaxel in Ovarian Cancer : focus on interindividual differences in pharmacodynamics and pharmacokinetics

    Author : Henrik Green; Curt Peterson; Peter Söderkvist; Per Rosenberg; Håkan Melhus; Linköpings universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; pharmacogenetics; paclitaxel; LC MS; Taxol; ovarian cancer; pharmacokinetics; CYP2C8; CYP3A4; ABCB1; äggstockscancer; paklitaxel; Clinical pharmacology; Klinisk farmakologi;

    Abstract : Ovarian cancer is one of the most common female cancer diseases in the world today and in Sweden more than 800 new cases are diagnosed every year. The standard treatment consists of chemotherapy with paclitaxel in combination with carboplatin after initial cytoreductive surgery. READ MORE

  2. 2. Intestinal Permeability and Presystemic Extraction of Fexofenadine and R/S-verapamil

    Author : Christer Tannergren; Hans Lennernäs; Dirk KF Meijer; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Biopharmacy; Bioavailability; Fexofenadine; Verapamil; P-glycoprotein; CYP3A4; OATP; Enantioselective; Permeability; Caco-2; Intestinal perfusion; Biofarmaci; Biopharmacy; Biofarmaci;

    Abstract : The main objective of this thesis was to investigate the in vivo relevance of membrane transporters and cytochrome P450 (CYP) 3A4-mediated metabolism in the intestine and liver for the bioavailability of drugs in humans after oral administration.In the first part of the thesis, the main transport mechanisms involved in the intestinal absorption and bioavailability were investigated for fexofenadine, a minimally metabolized drug, which is a substrate for P-glycoprotein (P-gp) and members of organic anion transporting polypeptide (OATP) family. 3. 3. Qualitative and Quantitative Assessment of Cytochromes P450 mRNA in Human : Studies in the Liver, Blood and Gastrointestinal Mucosa

    Author : Mari Thörn; Lars Lööf; Anders Rane; Ellen Vinge; Uppsala universitet; []
    Keywords : Medical sciences; cytochrome P450; CYP1A2; CYP1B1; CYP2E1; CYP3A4; CYP3A5; P-gp; gene expression; RT-PCR; liver; blood; gastrointestinal mucosa; MEDICIN OCH VÅRD; MEDICINE; MEDICIN;

    Abstract : Drugs and other foreign compounds must often be metabolised before they can be excreted from the body. One enzyme system that is responsible for this is the cytochrome P450 gene family (CYP). READ MORE

  4. 4. Small non-coding RNAs in cancer

    Author : Helena Persson; Bröstcancer-genetik; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; microRNA; non-coding RNA; breast cancer; CYP3A4; bladder cancer;

    Abstract : The complex genetics of cancer, which allows tumours to grow and spread while bypassing the control circuitry of the cell, involves dysregulation of the genome, transcriptome and proteome. The importance of protein-coding genes such as growth factors and hormone receptors is well established. READ MORE

  5. 5. In vivo Pharmacokinetic Interactions of Finasteride and Identification of Novel Metabolites

    Author : Anna Lundahl; Hans Lennernäs; Dennis A Smith; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; drug-drug interactions; herb-drug interactions; pharmacokinetics; metabolism; CYP3A4; hydroxylation; glucuronidation; UGT enzymes; mass spectrometry; biliary excretion; urinary excretion; finasteride; PHARMACY; FARMACI;

    Abstract : The general aim of this thesis was to improve the understanding of the in vivo pharmacokinetics and, in particular, the metabolism of finasteride, a 5α-reductase inhibitor used in the treatment of enlarged prostate glands and male pattern baldness. CYP3A4 has been identified as the major enzyme involved in the sequential metabolism of finasteride to ω-OH finasteride (M1) and ω-COOH finasteride (M3). READ MORE
