  1. 1. Exploitation of interactions with the neonatal Fc receptor to manipulate biological half-lives for therapeutic applications

    Author : Shengze Yu; Torbjörn Gräslund; Mats Ohlin; KTH; []
    Keywords : Glioblastoma Multiforme GBM ; Prolacin Prl ; Prolacin receptor PrlR ; Prolactin antagonist PrlRA ; Albumin binding domain ABD ; serum albumin SA ; half-life extension; FcRn; Affibody; human growth hormone hGH ; immunoglobulin G IgG ; blocking; autoimmune disease; Bioteknologi; Biotechnology;

    Abstract : Protein engineering provides powerful tools to create useful proteins with desired properties. In this thesis, rational design principles have been used for development of fusion proteins that can interact with the neonatal Fc receptor (FcRn) for potential medical applications. FcRn is widely expressed in the human body. 2. 2. Rational and Combinatorial Engineering of Affinity Proteins Towards Therapeutical Applications

    Author : Johan Seijsing; Torbjörn Gräslund; Uhlén Mathias; Inger Sandlie; KTH; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; neonatal Fc receptor; FcRn; affibody molecule; immunoglobulin G IgG ; serum albumin SA ; albumin binding domain ABD ; half-life extension; blocking; autoimmune disease; pH-dependence; GAD67; GAD-1; gene therapy; Parkinson’s disease; zinc finger based artificial transcription factor; Biotechnology; Bioteknologi;

    Abstract : Protein engineering has had an immense impact on the development of biological drugs, including replacement therapies with engineered versions of insulin or factor VIII to treat diabetes or bleeding disorders, and monoclonal antibodies to treat cancer and various other malignancies. Now, with the next generation of treatment modalities coming up, including monoclonal reagents based on alternative scaffolds, gene and cell therapies, the importance of protein engineering to tailor-make these treatments is likely to increase further. 3. 3. Development of the gastrointestinal tract in young mammals : Effects of enteral provocation with protease or phytohaemagglutinin in neonatal rats

    Author : Ester Arevalo Sureda; Funktionell zoologi; []
    Keywords : gut; Intestine; Pancreas; development; precocious; protease; PHA; enterocytes; permeability; endocytosis; IgG; FcRn; Blimp1; T-lymphocyte; passive immunity; altricial; neonatal; suckling; athymic; rat; immunohistochemistry IHC ;

    Abstract : The rat, as an altritial species, is born with an immature gastrointestinal tract and intestinal barrier function, which is highly absorptive to milk-borne bioactive molecules that can pass undigested and reach the general circulation of the suckling newborn. This passage occurs by the neonatal-Fc-receptor (FcRn) binding and trancytosis of immunoglobulin G in the proximal small intestine (SI) and by the highly endocytic vacuolated enterocytes non-selectively in the distal SI. 4. 4. One-click away from higher contrast : Improvements to peripheral clearance for same-day immunoPET in Alzheimer’s disease

    Author : Eva Schlein; Dag Sehlin; Jordi Llop Roig; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Alzheimer’s disease AD ; bispecific antibody; Amyloid-β Aβ ; Receptor mediated transcytosis RMT ; Neonatal Fc receptor FcRn ; Blood-brain barrier BBB ; Inverse electron demand Diels−Alder reaction IEDDA; pre-targeting; tetrazine; bio-orthogonal; Fluorine-18; clearing agent;

    Abstract : The brain is a challenging target for antibody-based positron emission tomography (immunoPET) to image amyloid-beta (Aβ). Antibodies detect pathology with high sensitivity, but due to their size and biological half-life, they cause a high background radiation, if radiolabelled. READ MORE

  5. 5. Fc receptors and feedback regulation by antibodies

    Author : Mikael Karlsson; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Genetics; Mouse; B lymphocytes; Fc; receptors; IgG; IgE; FcgRI; FcgRII; FcgRIII; Fc e RI; CD23; Rh prophylaxis; Haemolytic disease of the newborn; Genetik; Clinical genetics; Klinisk genetik; Pathology; patologi;

    Abstract : The purpose of this investigation was to determine the role of Fc receptors for IgG (FcγRI, FcγRIIB, FcγRIII and FcRn) in immunoregulation. IgG is known to enhance the response to soluble antigens such as bovine serum albumin whereas the response to particulate antigens is suppressed. READ MORE
