  1. 1. Cooperative Reactive Sites in Designed Helix-Loop-Helix Polypeptide Catalysts

    Author : Jonas W. Nilsson; Paolo Scrimin; Linköpings universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES;

    Abstract : Designed polypeptides, which fold into helix-loop-helix motifs and dimerise to four-helix bundles have been used as scaffolds for the introduction of catalytic activity for the hydrolysis of p-nitrophenyl esters. The reactive sites were based on the reactivity of histidine, and two His residues separated by four residues in the sequence was shown to be the minimal reactive site for the efficient hydrolysis of nitrophenyl esters. 2. 2. Polypeptide-Based Nanoscale Materials

    Author : Daniel Aili; Bo Liedberg; Karin Enander; Molly Stevens; Linköpings universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Gold nanoparticle; polypeptide; helix-loop-helix; four-helix bundle; self-assembly; folding; Chemistry; Kemi;

    Abstract : Self-assembly has emerged as a promising technique for fabrication of novel hybrid materials and nanostructures. The work presented in this thesis has been focused on developing nanoscale materials based on synthetic de novo designed polypeptides. READ MORE

  3. 3. Protein-protein interactions in model systems : design, control of catalytic activity and biosensor applications

    Author : Johan Rydberg; Lars Baltzer; Joel Schneider; Linköpings universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Chemistry; Protein interactions; design; catalysis; biosensors; hybride materials; nanoparticles; Kemi; Chemistry; Kemi;

    Abstract : This thesis describes the design of polypeptides, unordered in the monomeric state but capable of folding into helix-loop-helix motifs and dimerise to form four-helix bundles. The goal of the design was to encode them with the capacity to form dimers highly selectively and the ability to carry out molecular functions in the folded state but not in the unordered state, and thus to establish a molecular link between recognition and function. 4. 4. Targeting Biological Systems by Organic Synthesis Methods - Cancer Cells and Proteins

    Author : Cecilia Winander; Lars Baltzer; Stefan Sjöberg; Jörgen Carlsson; Vladimir Tolmachev; Thomas Schrader; Uppsala universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Organic chemistry; carborane; doxorubicin; 125I; molecular recognition; four-helix bundle; polypeptide conjugate; affinity; chitinase; acetylcholine esterase; Organisk kemi;

    Abstract : This thesis describes the design and synthesis of molecules with potential roles in biomedicine, with an emphasis on molecular recognition in complex biological environments. The first chapter describes the synthesis and evaluation of compounds for use in nuclide therapy. READ MORE

  5. 5. NMR studies on calcium-induced conformational transitions in calmodulin

    Author : Johan Evenäs; Biofysikalisk kemi; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Physical chemistry; NMR; conformational exchange; calmodulin; calcium binding; Fysikalisk kemi;

    Abstract : Nuclear magnetic resonance (NMR) spectroscopy was used in order to investigate the relationships between structure, dynamics and calcium binding in the intracellular regulatory protein, calmodulin. Calmodulin consists of two similar domains, each binding two calcium ions with positive cooperativity. READ MORE
