    Abstract : In this thesis reductions of model and anticancer active Pt(IV) complexes with thiols and ascorbic acid and substitution reactions of cisplatin and model Pt(II) complexes with S-donor nucleophiles (rescue agents) have been investigated using stopped-flow and UV/VIS spectrophotometry. Glutathione (GSH) reduction of the anticancer active Pt(IV) compounds t- and c-[PtCl4(cha)(NH3)] (cha = cyclohexylamine), t-[PtCl4(thiazole)(NH3)] and c-[PtCl4(NH3)2] at 25 oC was observed to be very fast.     Abstract : Reactions of trans-dicyanodichloroaurate(III) trans-dibromodicyanoaurate(III), trans-diamminedichlorogold(III), cis-diammine(aqua)chloroplatinum(II), cis-ammineaqua-(cyclohexylamine)chloroplatinum(II), chloro(terpyridine)platinum(II) and chloro-(diethylenetriaminepalldium(II) with dimethyl sulfide, thionucleosides, thionucleotides and thione- and guanine-containing 17-mer oligonucleotides have been investigated by use of conventional and high-pressure stopped-flow spectrophotometry and HPLC technique as a function of ionic strength, pH, temperature and pressure in aqueous solutions. The mechanism and rate of oxidation of dimethyl sulfide by gold(III) complexes depends on the concentration of free halide. READ MORE

    Abstract : The kinetics and mechanism for substitution reactions on tetraaquapalladium(II) by an extended series of thioethers, a great number of carboxylic acids, and olefins have been studied by use of UV-VIS, stopped-flow, and high-pressure stopped-flow spectrophotometry and NMR spectroscopy.     Abstract : It is well documented that the antitumour activity of the Pt(II)-complex cisplatin, and similar anticancer active complexes, is a result of metalation of DNA, and thereby inhibition of cell proliferation. Since nucleic acids are polyanions at physiological pH, the rate of adduct formation with positively charged complex, as the hydrolyzed Pt(II) complexes studied in this work, is influenced by the ionic environment surrounding the polymer.
