  1. 1. The Role of Stat1 in Retinoic Acid-induced Myelomonocytic Differentiation of Human Leukemia Cells

    Author : Anna Dimberg; Ivo Touw; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Genetics; Stat1; ATRA; U-937; myeloid; differentiation; cell cycle; growth arrest; Genetik; Clinical genetics; Klinisk genetik; Pathology; patologi;

    Abstract : All-trans retinoic acid (ATRA), a biologically active metabolite of vitamin A, is a powerful inducer of terminal differentiation and growth arrest of several myeloid cell lines in vitro. Although the efficacy of ATRA as an anti-cancer drug has been demonstrated by the successful treatment of acute promyelocytic leukemia (APL), knowledge concerning the molecular mechanisms directing ATRA-induced differentiation and cell cycle arrest of myeloid cells is lacking. 2. 2. Expression profiling using manifold supports

    Author : Anette Hagberg; Joakim Lundeberg; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Genetics; mRNA; expression; minimal residual disease MRD ; chronic myeloid leukemia CML ; recombinant human erythropoietin r-HuEpo ; anemia; insulin dependent diabetes mellitus IDDM ; transplantation; Genetik; Clinical genetics; Klinisk genetik; Medical Genetics; medicinsk genetik;

    Abstract : Analyses of mRNA provides a condensed view of gene structure and quantitative analysis can reveal the induction of physiological or pathological gene expression programs. This thesis describes a new method for mRNA isolation, followed by sensitive real time detection via polymerase chain reaction (PCR), in order to quantitate transcripts of interest. READ MORE

  3. 3. Characterization of Genetic Abnormalities at Disease Progression of Chronic Myeloid Leukemia

    Author : Aikaterini Barbouti; Avdelningen för klinisk genetik; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; fluorescent in situ hybridization; low-copy repeats; isochromosome 17q; MSI2 HOXA9; ETV6 ABL1; BCR ABL1; chronic myeloid leukemia; blast crisis; molecular genetics; Clinical genetics; Klinisk genetik;

    Abstract : Chronic myeloid leukemia (CML) is a myeloproliferative disorder associated with the translocation t(9;22)(q34;q11) and progresses from a relatively indolent chronic phase (CP) to an accelerated phase (AP) and finally to the more aggressive blast crisis (BC). The general aim of this thesis was to identify and characterize genetic abnormalities occurring at disease progression of CML using molecular cytogenetic, molecular genetic, and bioinformatic analyses. 4. 4. Genotoxic effects of systemic chemotherapy in cancer patients, with special focus on the relation between MSI, LOH and development of secondary cancers

    Author : Nasir K. Kamat; Ulf Rannug; Giovanni Corso; Stockholms universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Chemotherapy; Genetic instability; Microsatellites instability; Loss of heterozygosity; Mismatch repair; molekylärgenetik; Molecular Genetics;

    Abstract : Systemic chemotherapy results in both phenotypic and genotypic side effects. Genotoxicity posed by chemotherapy is a major concern since it induces DNA damage and instability in the patients’ genome. READ MORE

  5. 5. Trisomies in Hematologic Malignancies

    Author : Kajsa Paulsson; Avdelningen för klinisk genetik; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Clinical genetics; Klinisk genetik; cytogenetik; Genetik; cytogenetics; Genetics; leukemia; chromosome aberration; trisomy;

    Abstract : Acquired clonal chromosome aberrations, closely associated with leukemogenesis, are found in many hematologic malignancies. Although the balanced rearrangements, such as translocations and inversions, are the ones most commonly thought of in the context of leukemias, gains of chromosomes ? e.g.. READ MORE
