  1. 1. Towards a New Generation of Anti-HIV Drugs : Interaction Kinetic Analysis of Enzyme Inhibitors Using SPR-biosensors

    Author : Malin Elinder; U. Helena Danielson; Maurizio Botta; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; drug discovery; fragment; screening; reverse transcriptase; microbicides; interaction analysis; NNRTIs; Biochemistry; Biokemi; Biokemi; Biochemistry;

    Abstract : As of today, there are 25 drugs approved for the treatment of HIV and AIDS. Nevertheless, HIV continues to infect and kill millions of people every year. Despite intensive research efforts, both a vaccine and a cure remain elusive and the long term efficacy of existing drugs is limited by the development of resistant HIV strains. READ MORE

  2. 2. Biosensor Studies of Ligand Interactions with Structurally Flexible Enzymes : Applications for Antiviral Drug Development

    Author : Matthis Geitmann; U. Helena Danielson; Marc H. V. van Regenmortel; Uppsala universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Biochemistry; SPR biosensor; HIV-1 reverse transcriptase; HCMV protease; interaction kinetics; drug discovery; non-nucleoside inhibitor; resistance; Biokemi; Biochemistry; Biokemi;

    Abstract : The use of a surface plasmon biosensor fills a missing link in kinetic studies of enzymes, since it measures directly the interaction between biomolecules and allows determination of parameters that are determined only indirectly in activity assays. The present thesis deals with kinetic and dynamic aspects of ligand binding to two viral enzymes: the human cytomegalovirus (HCMV) protease and the human immunodeficiency virus type 1 reverse transcriptase (HIV-1 RT). 3. 3. Reverse transcriptase assay for analysis of resistance to anti-HIV drugs and their mechanism of action

    Author : Xingwu Shao; Karolinska Institutet; Karolinska Institutet; []
    Keywords : HIV-1; RT; antiviral drugs; NRTls; NNRTls; drug resistance; viral load;

    Abstract : Reverse transcriptase (RT) is a viral enzyme and one of the main targets for drugs against human immunodeficiency virus (HIV). This work presents a set of sensitive non-radioactive RT assays were developed: standard RT inhibition assay (IC assay); RT binding inhibition assay; RTprotein detection assay and template-primer destruction assay or chain termination assay (CT assay). 4. 4. Assays for monitoring HIV therapy in low-middle income countries

    Author : Eva Agneskog; Karolinska Institutet; Karolinska Institutet; []
    Keywords : ;

    Abstract : The purpose of this thesis was to develop and/or evaluate assays for HIV viral load (VL) monitoring and HIV drug resistance testing of potential importance for research studies and clinical care in low- and middle income countries (LMICs). This was achieved using reverse transcriptase based (RT) assays and blood samples from Vietnamese and Swedish HIV-1 infected patients. READ MORE

  5. 5. Optimization of pediatrics antiretroviral treatment outcomes among HIV infected children in Ethiopia

    Author : Birkneh Tilahun Tadesse; Karolinska Institutet; Karolinska Institutet; []
    Keywords : ;

    Abstract : Ending the HIV/AIDS epidemic by 2030 is a global agenda. To meet this global goal, having safe and effective antiretroviral therapy is a key requirement. In Ethiopia, the safety and efficacy of combination antiretroviral therapy in HIV infected pediatric population is poorly studied. READ MORE
