  1. 1. Structural Studies of Human 5'-Nucleotidases

    Author : Karin Walldén; Pär Nordlund; Norbert Sträter; Stockholms universitet; []
    Keywords : TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; Nucleotidases; nucleotide; nucleoside; nucleoside analogue; Structural biochemistry; Strukturbiokemi; biokemi; Biochemistry;

    Abstract : 5’-Nucleotidases (5’NTs) are catabolic enzymes of the nucleotide metabolism. They catalyze dephosphorylation of deoxyribo- and ribonucleoside monophosphates and constitute an important control point in the regulation of intracellular nucleotide pools for the maintenance of correct DNA and RNA synthesis. 2. 2. Pharmacology and resistance mechanisms of nucleoside analogues and topoisomerase II interactive agents : studies on human leukemia cells with a focus on cross-resistance

    Author : Kourosh Lotfi; Rolf Larsson; Linköpings universitet; []
    Keywords : Nucleoside analogue; cross-resistance; multidrug resistance; deoxycytidine kinase; leukemia; anthracyclines; P-glycoprotein; MEDICINE; MEDICIN;

    Abstract : The purpose of this thesis was to elucidate mechanisms of action and resistance of clinically relevant nucleoside analogues and topoisomerase interactive agents in human leukemia cell lines and leukernia cells isolated from peripheral blood of leukemia patients. Interactions and cross-resistance patterns of these different cytotoxic drug families were also studied since these drugs are usually administrated in combination in the clinic. READ MORE

  3. 3. Distribution of antiviral nucleoside analogues into brain and skin

    Author : Natalia Borg; Karolinska Institutet; Karolinska Institutet; []
    Keywords : ;

    Abstract : The aim of this thesis was to study physiological and physico-chemical factors influencing the pharmacokinetics and distribution over the blood-brain barrier of antiviral nucleoside analogues in the rat, and to study the distribution to the human skin in vivo of an anti-herpetic nucleoside analogue. Microdialysis was used to sample the free (unbound) extracellular concentration of the nucleoside analogues from the brain tissue, muscle and blood in the rat and from the dermis of healthy human volunteers. 4. 4. Nucleoside phosphoramidates and related compounds : development of synthetic methods based on H-phosphonate chemistry

    Author : Inger Kers; Wojciech J. Stec; Stockholms universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Organic Chemistry; organisk kemi;

    Abstract : In this thesis, studies directed towards development of new synthetic methods for the preparation of nucleotide analogues of potential medical value as antisense, antigene or antiviral agents, are described.Two methods for synthesis of N3'->P5' and P3'->N5' phosphoramidate analogues with the nitrogen atom in bridging position of the internucleotide linkage was developed. 5. 5. Pathogenic mechanisms affecting mitochondrial DNA replication and transcription

    Author : Meenakshi Singh; Göteborgs universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; mtDNA; ribonucleotides; nucleoside analogues; POLRMT;

    Abstract : Mitochondria are cytoplasmic organelles fundamental to life and health. In mitochondria, energy from the food we eat is converted into adenosine triphosphate (ATP), which in turn is used as a source of chemical energy to drive a multitude of cellular reactions. READ MORE
