    Abstract : The embryonic mouse inner ear was used as a model with which to study ototoxicity and tissue interactions. The inner ear anlage can be explanted and cultured in vitro from about the 12th gestational day (gd), and will differentiate parallel with the inner ear developing in vivo until a time corresponding to birth (21st gd).     Abstract : The principal aims of the studies described in this thesis were to develop an in vitro model for studying acoustic overstimulation at the cellular level, to define the electrical and mechanical response characteristics of the perfused temporal bone preparation, and to investigate the effects of intense sound stimulation on the calcium levels of the hair cells in the intact hearing organ. In the in vitro model for acoustic overstimulation, isolated cochlear outer hair cells were subjected to a pressure jet emanating from a glass micropipette aimed at the cell body. READ MORE

    Abstract : Using an in vitro model of the guinea pig, the temporal bone preparation, cochlear physiology was studied. By perfusion of the scala tympani with oxygenated culture medium, the responses of the hearing organ to sound stimuli were maintained over several hours and the functional state was followed by monitoring the electrophysiological parameters cochlear microphonics (CM) and summating potential (SP).
