  1. 1. Macrolide Antibiotics in Bacterial Protein Synthesis

    Author : Martin Lovmar; Måns Ehrenberg; Tanel Tenson; Peter B. Moore; Uppsala universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Biochemistry; protein synthesis; macrolides; resistance; initiation of translation; non-competitive inhibitors; kinetics; translation accuracy; proof-reading; Biokemi; Biochemistry; Biokemi;

    Abstract : Macrolides are a large group of clinically relevant antibiotics that inhibit protein synthesis by binding to the large ribosomal subunit in the peptide exit tunnel, close to the peptidyl transferase center (PTC). We have shown that the peptide length of the resulting peptidyl-tRNA drop-off products is proportional to the distance between the PTC and the respective macrolide in the tunnel. READ MORE

  2. 2. Mechanisms of Adaptation to Deformylase Inhibitors

    Author : Anna Zorzet; Dan Andersson; Paul Rainey; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; antibiotic resistance; compensatory evolution; compensatory mutations; protein synthesis; initiation factor 2; initiator tRNA; formylation; peptide deformylase inhibitors; Bacteriology; Bakteriologi; Mikrobiologi; Microbiology; Biokemi; Biochemistry; Geography; Geografi;

    Abstract : Antibiotic resistance is a growing problem on a global scale. Increasing numbers of bacteria resistant toward one or multiple antibiotics could return us to the high mortality rates for infectious diseases of the pre-antibiotic era. READ MORE

  3. 3. Design, Synthesis and Characterization of Small Molecule Inhibitors and Small Molecule : Peptide Conjugates as Protein Actors

    Author : Jonas Nilsson; Lars Baltzer; Ingemar Kvarnström; Thomas Olsson; Linköpings universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Synthetic receptor; Peptide inhibitor conjugate; Peptide protein surface interactions; General Lead Discovery Library; Combinatorial Chemistry; Solid Phase Chemistry; Targeted Library; Thrombin inhibitor; Fluorescence; Surface Plasmon Resonance; Organic chemistry; Organisk kemi;

    Abstract : This thesis describes different aspects of protein interactions. Initially the function of peptides and their conjugates with small molecule inhibitors on the surface of Human Carbonic Anhydrase isoenzyme II (HCAII) is evaluated. READ MORE

  4. 4. Glycoconjugates : synthesis and investigation of carbohydrate-protein interactions

    Author : Sara Spjut; Mikael Elofsson; Ulf Ellervik; Umeå universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Glycoconjugates; Carbohydrates; Galactose; Glucose; Solid-phase synthesis; SPOS; Glycosylation; Gel-phase 19F NMR spectroscopy; Fluorinated linker; Carbohydrate array; Microtiter plates; Carbohydrate-protein interactions; carbamate linker; Fsec; Protecting group; Fluorinated protecting group; Multivalent glycoconjugates; Adenovirus; Ad37; Epidemic keratoconjunctivitis; EKC; Sialic acid; Adenovirus inhibitor; Trivalent; Tetravalent; X-ray crystal structure; Bioorganic chemistry; Bioorganisk kemi; organisk kemi; Organic Chemistry;

    Abstract : To study the functions of glycoconjugates in biological systems reliable and efficient protocols for glycoconjugate synthesis are needed. To reach this goal we have developed methods for solid-phase synthesis of glycoconjugates that can be monitored with gel-phase 19F spectroscopy using fluorinated linkers, building blocks, and protecting groups. 5. 5. Purine and Pyrazolopyrimidine Derivatives: Design and Synthesis of Chemical Tools for Biological Applications

    Author : David Bliman; Göteborgs universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; purine; pyrazolopyrimidine; caged compounds; protein kinases; protein-protein interactions; fluorescence; organic synthesis;

    Abstract : Purines can be found in a multitude of naturally occurring compounds with a range of functions. This thesis describes the design and synthesis of purines and structurally related pyrazolopyrimidine derivatives intended for biological applications. READ MORE
