  1. 1. Novel Pharmacometric Methods for Informed Tuberculosis Drug Development

    Author : Oskar Clewe; Ulrika S.H. Simonsson; Mats O. Karlsson; Piet van der Graaf; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; pharmacokinetics; pharmacodynamics; PKPD; pharmacometric; nonlinear mixed-effects models; multistate tuberculosis pharmacometric model; general pharmacodynamic interaction model; general pulmonary distribution model; tuberculosis; rifampicin; isoniazid; ethambutol; Pharmaceutical Science; Farmaceutisk vetenskap;

    Abstract : With approximately nine million new cases and the attributable cause of death of an estimated two millions people every year there is an urgent need for new and effective drugs and treatment regimens targeting tuberculosis. The tuberculosis drug development pathway is however not ideal, containing non-predictive model systems and unanswered questions that may increase the risk of failure during late-phase drug development. 2. 2. Pharmacometric Models for Antibody Drug Conjugates and Taxanes in HER2+ and HER2- Breast Cancer

    Author : Brendan Bender; Lena Friberg; Mats Karlsson; Jin Jin; Oscar della Pasqua; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; pharmacometric; PKPD; model; breast cancer; T-DM1; thrombocytopenia; hepatotoxiticy; HER2; Pharmacokinetics and Drug Therapy; Farmakokinetik och läkemedelsterapi;

    Abstract : In oncology, there is a need to optimize drug treatment for efficient eradication of tumors, minimization of adverse effects (AEs), and prolonging patient survival. Pharmacometric models can be developed to streamline information between drug development phases, describe and quantify response to treatment, and determine dose regimens that balance toxicity and efficacy. 3. 3. Improved pharmacometric model building techniques

    Author : Radojka Savic; Mats Karlsson; Siv Jönsson; Matt Hutmacher; Uppsala universitet; []
    Keywords : Model building; Absorption model; Transit compartment model; Nonparametric method; Extended grid method; Semiparametric; Distribution transformation; Shrinkage; Model diagnostics;

    Abstract : Pharmacometric modelling is an increasingly used method for analysing the outcome from clinical trials in drug development. The model building process is complex and involves testing, evaluating and diagnosing a range of plausible models aiming to make an adequate inference from the observed data and predictions for future studies and therapy. 4. 4. Pharmacometrics Modelling in Type 2 Diabetes Mellitus : Implications on Study Design and Diabetes Disease Progression

    Author : Siti Maisharah Sheikh Ghadzi; Maria C. Kjellsson; Mats O. Karlsson; Sandra Visser; Uppsala universitet; []

    Abstract : Pharmacometric modelling is widely used in many aspects related to type 2 diabetes mellitus (T2DM), for instance in the anti-diabetes drug development, and in quantifying the disease progression of T2DM.The aim of this thesis were to improve the design of early phase anti-diabetes drug development studies with the focus on the power to identify mechanism of drug action (MoA), and to characterize and quantify the progression from prediabetes to overt diabetes, both the natural progression and the progression with diet and exercise interventions, using pharmacometrics modelling. 5. 5. Study Design and Dose Regimen Evaluation of Antibiotics based on Pharmacokinetic and Pharmacodynamic Modelling

    Author : Anders Kristoffersson; Lena Friberg; Mats Karlsson; Pernilla Lagerbäck; Otto Cars; Stephan Schmidt; Uppsala universitet; []
    Keywords : pharmacometric; optimal design; pharmacokinetics; pharmacodynamics; PKPD; resistance; antibiotics; modeling; time-kill curve; colistin; meropenem; ciprofloxacin; non-linear mixed effects models; bayesian; Farmaceutisk vetenskap; Pharmaceutical Science;

    Abstract : Current excessive use and abuse of antibiotics has resulted in increasing bacterial resistance to common treatment options which is threatening to deprive us of a pillar of modern medicine. In this work methods to optimize the use of existing antibiotics and to help development of new antibiotics were developed and applied. READ MORE
