  1. 1. Drug Partitioning into Natural and Artificial Membranes : Data Applicable in Predictions of Drug Absorption

    Author : Caroline Engvall; Birgitta Tomkinson; Katarina Edwards; Sven Engström; Uppsala universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Biochemistry; Bilayer disk; Cholesterol; Drug partitioning; Drugs; Electrostatic effects; Immobilized liposome chromatography; Liposome; Membrane protein; Model membrane; Oligonucleotide-liposome complex; Phospholipid; Phospholipid bilayer; Proteoliposome; Surfactant; Biokemi; Biochemistry; Biokemi;

    Abstract : When drug molecules are passively absorbed through the cell membrane in the small intestine, the first key step is partitioning of the drug into the membrane. Partition data can therefore be used to predict drug absorption. READ MORE

  2. 2. Chromatographic Studies of Solute Interactions with Immobilized Red Blood Cells and Biomembranes

    Author : Ingo Gottschalk; Irving W. Wainer; Uppsala universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Biochemistry; Affinity; Binding; Biomembrane; Biotin; Chromatography; Cytochalasin B; Dissociation constant; Drug absorption; Equilibrium; Glucose; GLUT1; Immobilization; Immobilized biomembrane affinity chromatography; Immobilized liposome chromatography; Interaction; Liposome; Membrane protein; Membrane vesicle; Partitioning; Phospholipid bilayer; Proteoliposome; Quantitative; Red blood cell; Solute; Specific; Streptavidin; Wheat germ agglutinin; Biokemi; Biochemistry; Biokemi; Biokemi; Biochemistry;

    Abstract : Specific and non-specific interactions of solutes with immobilized biomembranes were studied using chromatographic methods. Liposomes, proteoliposomes and red blood cell (RBC) membrane vesicles were immobilized by a freeze-thawing procedure, whereas whole RBCs were adsorbed in the gel beds using electrostatic interaction, binding to wheat germ agglutinin (WGA) or the streptavidin-biotin interaction. Abstract : The human glucose transporter GLUT1 is abundant in red blood cells, the blood-brain barrier and epithelial cells, where it mediates the transport of the energy metabolite, glucose. In the present work some properties of GLUT1, including affinity binding of both substrates and inhibitors, transport rates as well as permeabilities of aromatic amino acids and drug-membrane interactions were analyzed by chromatographic methods. 4. 4. Quantitation, Purification and Reconstitution of the Red Blood Cell Glucose Transporter GLUT1

    Author : Shusheng Zuo; Bengt Mannervik; Bengt Persson; Uppsala universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Biochemistry; Cholesterol; Cytochalasin B; Glucose; GLUT1; Hummel and Dreyer analysis; Immobilization; PEG 5000 -DSPE; Biomembrane; Proteoliposome; Quantitative frontal affinity chromatography; Red blood cell streptavidin-biotin immobilization; Sulfhydryl affinity chromatography; The modified micro-Bradford CaPE assay; MALDI-ToF-MS; Biokemi; Biochemistry; Biokemi;

    Abstract : The human glucose transporter GLUT1 facilitates glucose to be accumulated on the other side of the cell membrane. The functional state of GLUT1 is uncertain due to diversity of the reports. In this thesis, the activity of red blood cell GLUT1 was extensively studied to further characterize this protein. 5. 5. Plant aquaporin regulation: Structural and functional studies using diffraction and scattering techniques

    Author : Michael Järvå; Göteborgs universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; X-ray Crystallography; Membrane protein; Plasma membrane intrinsic protein; Aquaporin; pH gating; mercury; proteoliposome assay; water transport; SANS; SAXS; WAXS; nanodisc; liposome;

    Abstract : Water is the basis for life as we know it. It is only logical then that all organisms have evolved specialized proteins, aquaporins, which regulate water flow across their membranes. Plants, which are immobile, depend more on their environment and also use water flows to move, to breathe, and to grow. READ MORE
