  2. 2. Multivariate spectroscopic methods for the analysis of solutions

    Author : Kent Wiberg; Sven Jacobsson; Lars Nørgaard; Stockholms universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Chemometrics; UV-Vis spectroscopy; Multivariate calibration; Lidocaine; Identity; Content; PLS; SIMCA; Non-column; Diode array UV spectroscopy; DAD; Control sample; High Capacity Analysis HCA ; Fluorescence spectroscopy; Albumin; Immunoglobulin G; HPLC-DAD; Prilocaine; Peak purity determination; PCA; PARAFAC; Partial separation; Curve resolution; Analytical chemistry; Analytisk kemi;

    Abstract : In this thesis some multivariate spectroscopic methods for the analysis of solutions are proposed. Spectroscopy and multivariate data analysis form a powerful combination for obtaining both quantitative and qualitative information and it is shown how spectroscopic techniques in combination with chemometric data evaluation can be used to obtain rapid, simple and efficient analytical methods. 3. 3. Biomarkers of internal exposure/dose : Methods to quantify adducts to protein and DNA by LC/MS studied with benzo[a]pyrene and isocyanates

    Author : Emelie Westberg; Margareta Törnqvist; Kimmo Peltonen; Stockholms universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; biomarker; PAH; serum albumin; adduct; DNA; isocyanate; miljökemi; Environmental Chemistry;

    Abstract : This thesis focuses on methods for quantification by liquid chromatography/mass spectrometry (LC/MS) of specific biomarkers for internal dose of chemicals which induce toxicity through their electrophilic reactivity. In vivo such compounds are short-lived, and could feasibly be measured as their reaction products (adducts) with biomacromolecules. READ MORE

    Abstract : Protein engineering has had an immense impact on the development of biological drugs, including replacement therapies with engineered versions of insulin or factor VIII to treat diabetes or bleeding disorders, and monoclonal antibodies to treat cancer and various other malignancies. Now, with the next generation of treatment modalities coming up, including monoclonal reagents based on alternative scaffolds, gene and cell therapies, the importance of protein engineering to tailor-make these treatments is likely to increase further.
