1. Immunoglobulin Gene Analysis in Different B cell Lymphomas : With Focus on Cellular Origin and Antigen Selection
Abstract : B cell lymphoma (BCL) comprises a biologically and clinically heterogeneous group of tumors deriving from different stages of B cell development. The immunoglobulin (Ig) variable heavy chain (VH) gene rearrangement is unique for each BCL and can be used to reveal cellular origin, to study signs of antigen selection and to quantify tumor cell load. 2. Analysis of Immunoglobulin Genes and Telomeres in B cell Lymphomas and Leukemias
Abstract : B cell lymphomas and leukemias are heterogeneous tumors with different cellular origins. Analysis of immunoglobulin (Ig) genes enables insight into the B cell progenitor, as Ig somatic hypermutation correlates with antigen-related B cell transit through the germinal center (GC). 3. Stereotyped B Cell Receptors in Chronic Lymphocytic Leukaemia : Implications for Antigen Selection in Leukemogenesis
Abstract : Biased immunoglobulin heavy variable (IGHV) gene usage and distinctive B-cell receptor (BCR) features have been reported in chronic lymphocytic leukaemia (CLL), which may reflect clonal selection by antigens during disease development. Furthermore, the IGHV gene mutation status distinguishes two clinical entities of CLL, where patients with unmutated IGHV genes have an inferior prognosis compared to those with mutated IGHV genes. 4. Role of DNA repair in class switch recombination and somatic hypermutation
Abstract : Class switch recombination (CSR) and somatic hypermutation (SHM), occurring in the germinal center, are two important processes for B cell development. Both are initiated by activation-induced cytidine deaminase (AID), through deamination of the C residues in the variable and switch regions of the immunoglobulin locus, resulting in either in single stranded or double stranded DNA breaks. 5. Studies of molecular events during peripheral B cell differentiation
Abstract : This thesis focuses on molecular changes occuring during terminal B cell differentiation as well as on somatic hypermutation and the generation and maintenance of B cell memory. During T cell dependent immune responses the average affinity for the eliciting antigen increases with time. READ MORE
