  1. 1. Fluorescence fluctuation studies of biomolecular interactions in solutions, biomembranes and live cells

    Author : Volodymyr Chmyrov; Jerker Widengren; Antoine Delon; KTH; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; FCS; FCCS; Isomerization; TRAST; NMR; FRET; biomombrane; fluidity; Biological Physics; Biologisk fysik;

    Abstract : Fluorescence spectroscopy and imaging have a very broad spectrum of applicationswithin the life sciences, in particular for detection and characterization ofbiomolecular dynamics and interactions in different environments. This thesis comprisesprojects that strive to further expand the information content extracted fromthe detected fluorescence, leading to sensitive readout parameters for studies ofbiomolecular dynamics and interactions. 2. 2. Transient State Fluorescence Microscopy - method development and biological applications : Exploiting the dark states of fluorophores to measure oxygen concentrations, redox state, Förster resonance energy transfer and membrane viscosity

    Author : Thiemo Spielmann; Jerker Widengren; Karsten König; KTH; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Transient States imaging TRAST ; Triplet State imaging; fluorescence microscopy; modulated excitation; triplet state; radical state; trans-cis isomerisation;

    Abstract : Due to their long lifetime, triplet, redox and other transient states of fluorophores are highly sensitive to the micro-environment. Imaging their spatial distribution in biological samples can therefore help answer interesting questions about the metabolism, molecular interactions and dynamics in living cells. READ MORE

  3. 3. Concepts and biomedical applications of excitation-modulated transient state monitoring of fluorescence emitters

    Author : Baris Demirbay; Jerker Widengren; Yves Mely; KTH; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Biological and Biomedical Physics; Biologisk och biomedicinsk fysik;

    Abstract : Fluorescence methods have developed very strongly in the last decade, allowing single- molecule detection sensitivity, high specificity, high time- and spatial resolution, as well as high readout speeds. Together, this makes fluorescencea very central readout modality for biomolecular and cellular studies. READ MORE

  4. 4. Advanced fluorescence-based fluctuation methods for biosensing

    Author : Elin Sandberg; Jerker Widengren; Victoria Birkedal; KTH; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Physics; Fysik;

    Abstract : In addition to use traditional fluorescence parameters for biological studies,the main focus of this thesis is on the monitoring of photoinduced fluorophoretransient states. These dark states, such as the triplet state, redox states andphotoisomers that are formed upon excitation, are long-lived in comparisonto the fluorescent singlet state and therefor more sensitive to the surroundingenvironment and their sensitivity are to biological relevant conditions. 5. 5. Fluorescence-based Transient State Monitoring for biomolecular, cellular and label-free studies

    Author : Johan Tornmalm; Jerker Widengren; Maria Garcia Parajo; KTH; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Biological Physics; Biologisk fysik; Fysik; Physics;

    Abstract : Fluorophore blinking dynamics are highly sensitive to the local environment and can be used as an additional readout parameter to increase the information gained from existing fluorescence techniques.The origin of these blinking patterns are photophysical transitions to and from a manifold of non-luminescent states.
