  1. 1. Proximity Extension Assay : a universal tool for proteomics in precision medicine

    Author : Phathutshedzo M. Muthelo; Ulf Landegren; Jochen Schwenk; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Proximity Extension Assay; Precision medicine; Assay development; Biology with specialization in Molecular Biology; Biologi med inriktning mot molekylärbiologi;

    Abstract : Precision medicine aims to utilise biomarkers to ensure the right treatment is given to the right patient at the right time. In pursuit of this goal, sensitive protein detection is essential to improve opportunities for discovery, verification, validation and development of valuable protein biomarkers. READ MORE

  2. 2. Antibody based plasma protein profiling

    Author : Ulrika Qundos; Jochen M. Schwenk; Peter Nilsson; Mathias Uhlén; Ulrike Korf; KTH; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; protein profiling; plasma; antibody; affinity proteomics; biomark- ers; multiplex; assay development;

    Abstract : This thesis is about protein profiling in serum and plasma using antibody suspension bead arrays for the analysis of biobanked samples and in the context of prostate cancer biomarker discovery. The influence of sample preparation methods on antibody based protein profiles were investigated (Papers I-III) and a prostate cancer candidate biomarker identified and verified (Papers III-V). 3. 3. Antibodies and Antibody Mimics in Binding Assays

    Author : Ioana Wärnmark; Tillämpad biokemi; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Polymerteknik; biopolymers; ELISA; antibody; molecularly imprinted polymer; Biochemistry; Biokemi; Metabolism; metabolism; Biokemisk teknik; Polymer technology; Biochemical technology;

    Abstract : This thesis presents development of new analytical methods using antibodies and molecularly imprinted polymers (MIPs) in binding assays: 1) The high specificity of antibodies and the wide variety of efficient labels make immunoassay one of the most popular analytical techniques today. Enzyme-linked immunosorbent assay (ELISA) is most widely used, since the enzyme label can be used as a catalyst to provide very high signal amplification. READ MORE

  4. 4. Production of polyclonal antibodies in rabbits and chickens immunised with human immunoglobulin G : A study of the utility of egg yolk antibody production as a substitute for rabbit serum antibody production

    Author : Lise Svendsen Bollen; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Laboratory animals; antibody production; egg yolk; adjuvants; antibody response; avidity; Försöksdjursvetenskap; Laboratory animal science; Försöksdjursvetenskap; zoologisk utvecklingsbiologi; Animal Development;

    Abstract : The utility of chicken egg yolk antibodies as an alternative to rabbit antisera was studied by comparing antibody and avidity development in immunised animals (15 rabbits and 30 chickens) during a one-year immunisation scheme. Human IgG was used as the model antigen and the efficacy of three adjuvants, Freund's Complete Adjuvant, Freund's Incomplete Adjuvant and Hunter's TiterMax, was compared. READ MORE

  5. 5. Advanced molecular tools for diagnostic analyses of RNA and antibodies in situ and in solution

    Author : Mengqi Wang; Ulf Landegren; Anders Ståhlberg; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Homogenous serological assay; PCR-based antibody detection; Proximity extension assay; SARS-CoV-2 antibody; Extracellular vesicles; Proximity barcoding assay; in situ superRCA; mutant transcript;

    Abstract : Advanced molecular diagnostics uses in vitro biological assays to detect nucleic acids or proteins even in low concentrations across samples, allowing for the identification of biomarkers, monitoring the course of the disease over time, and selection of appropriate therapy. In this thesis, I focus on development and early applications of several molecular tools of expected value in research, and eventually also clinically. READ MORE
