  1. 1. Blood-Brain Barrier Transport : Investigation of Active Efflux using Positron Emission Tomography and Modelling Studies

    Author : Stina Syvänen; Margareta Hammarlund-Udenaes; Mats Bergström; Gunnar Blomquist; Jashvant Unadkat; Uppsala universitet; []
    Keywords : Pharmaceutical biosciences; pharmacokinetics; P-glycoprotein; blood-brain barrier; modelling; PET; active efflux; species differences; [11C]verapamil; drug development; Farmaceutisk biovetenskap;

    Abstract : This thesis examines the transport of exogenous molecules across the blood-brain barrier (BBB), focusing on active efflux, using positron emission tomography (PET), computer simulation and modelling. P-glycoprotein (P-gp) inhibition was studied using [11C]verapamil and [11C]hydroxyurea was investigated as a new marker for active efflux transport. READ MORE

  2. 2. Quantitative Aspects of Nanodelivery Across the Blood-Brain Barrier : Exemplified with the Opioid Peptide DAMGO

    Author : Annika Lindqvist; Margareta Hammarlund-Udenaes; Markus Fridén; William Elmquist; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; blood-brain barrier; liposomes; nanocarriers; brain delivery; pharmacokinetics; modeling and simulation; microdialysis; opioid peptide; DAMGO; LC-MS MS; Farmaceutisk vetenskap; Pharmaceutical Science;

    Abstract : The use of nanocarriers is an intriguing approach in the development of efficacious treatment for brain disorders. The aim of the conducted research was to evaluate and quantify the impact of a liposomal nanocarrier formulation on the brain drug delivery. READ MORE

  3. 3. Pericyte Influence on Drug Delivery Across the Blood-Brain Barrier : Implications for Therapy of Neurodegenerative Diseases

    Author : Nebojsa Mihajlica; Margareta Hammarlund-Udenaes; Christer Betsholtz; William Elmquist; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Blood-brain barrier; pericytes; pericyte-deficiency; pharmacokinetics; small-molecular drugs; drug distribution; transporters; P-glycoprotein; tyrosine kinase inhibitors; Farmakokinetik och läkemedelsterapi; Pharmacokinetics and Drug Therapy;

    Abstract : The blood-brain barrier (BBB) represents a complex interface between the brain parenchyma and systemic blood circulation, strictly controlling exchange of substances between the two sites. Pericytes are mural cells located on the abluminal membrane of the brain endothelium, involved in BBB formation and maintenance. READ MORE

  4. 4. Developmental Aspects of Drug Transport Across the Blood-Brain Barrier

    Author : Jörgen Bengtsson; Margareta Hammarlund-Udenaes; Ann-Cathrin Engwall; Sven Björkman; Elizabeth C.M. de Lange; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Blood-brain barrier; development; active transport; tight junction proteins; microdialysis; recovery; morphine; nitrofurantoin; Bcrp; microarray; real-time PCR; in vitro; in vivo; LC-MS MS; Biopharmacy; Biofarmaci; Pharmacokinetics and Drug Therapy; Farmakokinetik och läkemedelsterapi;

    Abstract : The developmental aspect of drug transport across the blood-brain barrier (BBB) was investigated. Microdialysis was used to study unbound morphine BBB transport at different ages in sheep. An in vitro study was performed to find differentially expressed genes in brain capillary-rich fractions of the brain in rats of different ages. READ MORE

  5. 5. In Vivo Active Drug Uptake and Efflux at the Blood-Brain Barrier : With Focus on Drug Transport Interactions

    Author : Muhammad Waqas Sadiq; Margareta Hammarlund-Udenaes; Sven Björkman; N. Joan Abbott; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Blood-brain barrier; Active uptake transport; Microdialysis; P-glycoprotein; Drug interactions; Species differences; Pharmacokinetics; Pharmacodynamics; NONMEM; Pharmaceutical Science; Farmaceutisk vetenskap;

    Abstract : The blood-brain barrier (BBB) controls the movement of substances into and out of the brain. The tight junctions between endothelial cells and energy dependent transporters in the BBB influence rate and extent of drug distribution to the brain. READ MORE
