  1. 1. Fibroblast growth factor-23 and Klotho in bone/mineral and parathyroid disorders

    Author : Tijana Krajisnik; Tobias E. Larsson; Kenneth B. Jonsson; Östen Ljunggren; Peyman Björklund; Gunnar Sterner; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; calcitriol; chronic kidney disease; CKD; chronic renal failure; fibroblast growth factor 23; fibroblast growth factor-23; FGF23; FGF-23; GalNac-T3; GALNT3; GFR; hyperostosis-hyperphosphatemia syndrome; HHS; Klotho; parathyroid hormone; PTH; hyperparathyroidism; pHPT; sHPT; uremic; vitamin D3; Endocrinology; Endokrinologi; Internal Medicine; invärtesmedicin;

    Abstract : Fibroblast growth factor-23 (FGF23) is a novel, bone-produced hormone that regulates renal phosphate (Pi) reabsorption and calcitriol metabolism. Disorders of mineral and bone metabolism, such as autosomal dominant hypophosphatemic rickets (ADHR) and hyperostosis-hyperphosphatemia syndrome (HHS), witness the importance of well-balanced serum levels of FGF23. 2. 2. Enzymatic Regulation of Steroidogenesis and Nuclear Receptor Activation : Special Focus on Vitamin D and Sex Hormones

    Author : Johan Lundqvist; Kjell Wikvall; Maria Norlin; Magnus Ingelman-Sundberg; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; adrenal steroidogenesis; CYP7B1; vitamin D; calcitriol; enzymatic regulation; transcriptional regulation; CYP21A2; aromatase; sex hormone; estrogen; androgen; nuclear receptor; Pharmaceutical biochemistry; Farmaceutisk biokemi; Farmaceutisk biokemi; Pharmaceutical Biochemistry;

    Abstract : Enzyme-catalyzed reactions are important to regulate steroidogenesis and nuclear receptor activation. The present investigation examines the role of steroid metabolism catalyzed by CYP7B1 for regulation of hormone receptor activation and the effects of vitamin D on enzymatic regulation of steroidogenesis. 3. 3. Regulation of Vitamin D 25-hydroxylases : Effects of Vitamin D Metabolites and Pharmaceutical Compounds on the Bioactivation of Vitamin D

    Author : Maria Ellfolk; Kjell Wikvall; Maria Norlin; Olavi Pelkonen; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; CYP2D25; vitamin D 25-hydroxylase; transcriptional regulation; vitamin D; CYP2R1; CYP2J2; CYP27A1; phenobarbital; efavirenz; fibroblast; LNCaP; Pharmaceutical biochemistry; Farmaceutisk biokemi;

    Abstract : A 700bp portion of the promoter of CYP2D25, the porcine microsomal vitamin D 25-hydroxylase was isolated and sequenced. The computer analysis of the sequence revealed the existence of a putative VDRE at 220 bp upstream of the transcription start site. READ MORE

  4. 4. The role of fibroblast growth factor-23 in chronic kidney disease-mineral and bone disorder

    Author : Majd A. I. Mirza; Tobias Larsson; Tilman Drüeke; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; FGF23; FGF-23; CKD; CKD-MBD; cardiovascular disease; vascular function; atherosclerosis; hypertrophy; LVMI; LVH; fractures; bone; fat mass; obesity; apolipoproteins; cholesterol; lipids; Kidney diseases; Njursjukdomar; Cardiovascular medicine; Kardiovaskulär medicin; Medicinsk vetenskap; Medical Science;

    Abstract : Fibroblast growth factor-23 (FGF23) was initially identified as the causative factor of autosomal dominant hypophosphatemic rickets. Further studies confirmed that FGF23 is predominantly expressed in the osteocytes and osteoblasts of bone and that circulating FGF23 acts on the kidney to inhibit renal phosphate reabsorption and 1,25(OH)2D3 hydroxylation. 5. 5. Cytochrome P450 in the rat brain : characterization and regulation

    Author : Eva Hedlund; Karolinska Institutet; Karolinska Institutet; []
    Keywords : ;

    Abstract : Cytochrome P450s (CYPs) are key enzymes in the metabolism of cholesterol and fatty acids and their derivatives in the body. In the adrenals and gonads these enzymes are involved in steroid hormone biosynthesis, in the liver in bile acid synthesis, in the kidney in synthesis of the calcitropic hormone, calcitriol, and in the cardiovascular system in synthesis of thromboxane A2, a potent inducer of vasoconstriction and platelet aggregation.
