  1. 1. Amphiphilic Peptide Interactions with Complex Biological Membranes : Effect of peptide properties on antimicrobial and anti-inflammatory effects

    Author : Shalini Singh; Martin Malmsten; Per Hansson; Debora Berti; Uppsala universitet; []
    Keywords : LPS; Antimicrobial; Peptide; Inflammation; Infections; Liposome; Binding; PEG; Farmaceutisk fysikalisk kemi; Pharmaceutical Physical Chemistry;

    Abstract : With increasing problem of resistance development in bacteria against conventional antibiotics, as well as problems associated with diseases either triggered or enhanced by infection, there is an urgent need to identify new types of effective therapeutics for the treatment of infectious diseases and its consequences. Antimicrobial and anti-inflammatory peptides have attracted considerable interest as potential new antibiotics in this context. READ MORE

  2. 2. Evolution and Binding Mechanisms of Intrinsically Disordered Proteins

    Author : Elin Karlsson; Per Jemth; Kaare Teilum; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Intrinsically disordered proteins; Protein evolution; Binding kinetics; Binding mechanisms; Biophysical characterisation; Medicinsk biokemi; Medical Biochemistry;

    Abstract : Intrinsically disordered proteins (IDPs) make up a considerable fraction of the proteome in eukaryotic organisms. These proteins often act as hubs in interaction networks, harbouring multiple interaction with other proteins, and thus evolution has to walk a tightrope to accommodate new interactions while maintaining the previously established interactions. 3. 3. DNA Interactions with Chiral Polyaza-aromatic Ruthenium(II) Complexes

    Author : Per Lincoln; Chalmers tekniska högskola; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; phenanthroline; binding geometry; binding isotherm; DNA; dppz; linear dichroism; intercalation; ruthenium; cooperative binding;

    Abstract : The binding to DNA of a series of chiral ruthenium complexes is studied with optical spectroscopy techniques, including isotropic absorption, linear dichroism (LD) and emission anisotropy as well as steady-state and time-resolved luminescence. The thesis focuses on the ìlight-switchî complex [Ru(phen)2dppz]2+ which binds to DNA by intercalation and luminesces when bound to DNA but not in pure buffer solution (phen=1,10-phenanthroline; dppz=dipyrido[3,2-a:2',3'-c]phenazine). 4. 4. C4b-binding protein: Identification of binding sites and a possible function of the interaction with protein S

    Author : Joanna Webb; Malmö Klinisk kemi; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Clinical chemistry; apoptosis; binding sites; complement; C4b-binding protein; protein S; Klinisk kemi;

    Abstract : The subject of this thesis is plasma protein C4b-binding protein (C4BP). C4BP is an important regulator of the classical pathway of the complement system, a cascade-like system comprised of over 35 proteins, which partakes in the defence against micro-organisms and is involved of clearance of immune-complexes and apoptotic cells. 5. 5. NMR structral studies on ruthenium complexes binding to DNA

    Author : Lisha Wu; Chalmers tekniska högskola; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; NMR; DNA binding; Ru-complex dimer; CD; fluorescence; molecular dynamics simulation;

    Abstract : The Ru-complex Λ,Λ-[bidppz(bipy)4Ru2]4+ (Λ,Λ-B) exhibits multiple modes of interactions with the palindromic DNA dodecamer [d(CGCGAATTCGCG)]2, however excluding any type of intercalation as shown by both optical spectroscopy and NMR. NMR observation of Λ,Λ-B titration into a DNA solution results in both a transient binding, with observation of fast exchange for protons located near the ends of the DNA duplex, and a more stable interaction in the DNA minor groove indicated by a slow exchange mechanism and binding sites for G2, G4 and A5 of the same DNA strand.
