1. Development of benzoquinoquinoxaline derivatives as triplex-specific probes : Recognition of DNA structures at repeats sequences
Abstract : Repeat sequences are associated with several human diseases, such as Friedreich’s ataxia, polycystic kidney disease and cancer. These sequences can form non-B-DNA structures, including triplex (H-DNA) DNA, and are associated with genomic instability and altered gene expression. READ MORE

2. Structural Adaptation of DNA to Small Ligands and Recombination Proteins
Abstract : In order to contribute to the understanding of mechanisms of recognition in nucleic acid systems, the interaction of DNA with a variety of ligands, representing different cases of adaptation of the double-helix structure, was studied using flow linear dichroism (LD), circular dichroism (CD), fluorescence and absorption spectroscopic techniques. A chiral hydrophobic metal complex, [Ru(dpphen)3]2+ was examined, since this had been reported to be the most efficient molecule to discriminate DNA handedness. READ MORE

3. Nucleic Acids as Drug Targets - Influence of Platinum Complexes on Duplex Stability and Binding Kinetics
Abstract : Gaining a detailed understanding of how human cells are affected by metal-based drugs is crucial, since these drugs have shown great potential in cancer treatment. Cisplatin is used to treat many different types of cancer, e.g. testicular, ovarian, melanoma, bladder, non-small cell lung cancer, small cell lung cancer, myelomas and lymphomas. READ MORE

4. Functional and structural properties of eukaryotic DNA polymerase epsilon
Abstract : In eukaryotes there are three DNA polymerases which are essential for the replication of chromosomal DNA: DNA polymerase alpha (Pol alpha), DNA polymerase delta (Pol delta) and DNA polymerase epsilon (Pol epsilon). In vitro studies of viral DNA replication showed that Pol alpha and Pol delta are sufficient for DNA replication on both leading and lagging DNA strands, thus leaving the function of Pol epsilon unknown. 5. Nucleic Acids as Drug Targets -Interactions of Platinum Complexes, Polyamines, and siRNA with DNA and RNA Models
Abstract : Drugs targeting nucleic acids were studied in DNA and RNA model systems. The effects were investigated with respect to structural and thermodynamic influence, kinetics, and biological function. The drugs employed in this thesis were anticancer active platinum(II) complexes, polyamine derivatives, and siRNA. READ MORE
