1. Intermediate State Stabilisation in Enzyme Catalysis Structural studies of two serine proteases and a photosynthetic reaction centre
Abstract : Enzymes play a key role in every organisms life. They catalyse chemical reactions and may also respond to external stimulus. The reoccurring theme of this thesis is how biologically relevant information can be gleaned from X-ray structures of the enzymes. READ MORE

2. Specification of Dorsal and Intermediate Telencephalic Character
Abstract : The telencephalon is the most highly evolved region of the vertebrate central nervous system (CNS). The major structures of the telencephalon - the cortex and basal ganglia – derive from the dorsally positioned pallium and the ventrally positioned subpallium, respectively. READ MORE

3. Molecular Simulation of Enzyme Catalysis and Inhibition
Abstract : The reaction mechanisms for the hemoglobin degrading enzymes in the Plasmodium falciparum malaria parasite, plasmepsin II (Plm II) and histo-aspartic protease (HAP), have been analyzed by molecular simulations. The reaction free energy profiles, calculated by the empirical valence bond (EVB) method in combination with molecular dynamics (MD) and free energy perturbation (FEP) simulations are in good agreement with experimental data. 4. Theoretical modeling of metal- and enzyme catalyzed transformations
Abstract : This thesis is focused on describing and predicting catalytic reactions. The major part of the work is based on density functional theory (DFT). In some cases where the size of the investigated system precluded the use of more accurate methods molecular dynamics was employed. READ MORE

5. Catalysis and Site-Specific Modification of Glutathione Transferases Enabled by Rational Design
Abstract : This thesis describes the rational design of a novel enzyme, a thiolester hydrolase, derived from human glutathione transferase (GST) A1-1 by the introduction of a single histidine residue. The first section of the thesis describes the design and the determination of the reaction mechanism. READ MORE
