  1. 1. In vivo Pharmacokinetic Interactions of Finasteride and Identification of Novel Metabolites

    Author : Anna Lundahl; Hans Lennernäs; Dennis A Smith; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; drug-drug interactions; herb-drug interactions; pharmacokinetics; metabolism; CYP3A4; hydroxylation; glucuronidation; UGT enzymes; mass spectrometry; biliary excretion; urinary excretion; finasteride; PHARMACY; FARMACI;

    Abstract : The general aim of this thesis was to improve the understanding of the in vivo pharmacokinetics and, in particular, the metabolism of finasteride, a 5α-reductase inhibitor used in the treatment of enlarged prostate glands and male pattern baldness. CYP3A4 has been identified as the major enzyme involved in the sequential metabolism of finasteride to ω-OH finasteride (M1) and ω-COOH finasteride (M3). READ MORE

  2. 2. Reducing the morbidity of transurethral resection of the prostate based on patient selection, fluid absorption, and blood loss

    Author : Lars Sandfeldt; Karolinska Institutet; Karolinska Institutet; []
    Keywords : Absorption; blood loss; surgical; cardiovascular disease; electrocardiography; ethanol; finasteride; glycine; hemodynamics; intracranial pressure; mannitol; myocardium; personality; prostatectomy; prostatic hyperplasia; risk factors; ~urethral resection of prostate; ultrasonography; urodynamics.;

    Abstract : A higher long-term mortality due to myocardial infarction after TURP, compared to open surgery, irrigating fluid absorption and blood loss, prompted this investigation of whether prostate size has any correlation with cardiovascular risk factors, whether two common fluids differ in terms of harmful effects, and whether premedication with finasteride may reduce the blood loss during TURP. Paper I; Fifty-two patients were studied before the treatment of their bladder outlet obstruction. READ MORE

  3. 3. Clinical pharmacokinetic/pharmacodynamic modelling of 5a-reductase inhibitors for the treatment of benign prostatic hyperplasia

    Author : Per Olsson Gisleskog; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Pharmacy; FARMACI; PHARMACY; FARMACI; Biopharmaceutics; biofarmaci;

    Abstract : Dihydrotestosterone (DHT), a potent androgen necessary for the development of benign prostatic hyperplasia (BPH) is formed from testosterone by the enzyme 5α-reductase, of which there are two types. The irreversible 5α-reductase inhibitors dutasteride, which inhibits both isozymes, and finasteride, which inhibits only one, are intended for the treatment of BPH. READ MORE

  4. 4. Role of estrogen receptor beta in mouse ventral prostate with relation to human prostatic diseases

    Author : Otabek Imamov; Karolinska Institutet; Karolinska Institutet; []
    Keywords : ;

    Abstract : In this thesis we investigate role of estrogen receptor beta (ERbeta) in development and functioning of mouse ventral prostate, using it as a model for human extrapolations. ERbeta was discovered in 1995 and is a member of a nuclear receptors superfamily, acting as a transcription factor for steroid hormone signaling. READ MORE

  5. 5. Prostate cancer : aspects of screening and prognostic factors

    Author : Anders Kjellman; Karolinska Institutet; Karolinska Institutet; []
    Keywords : ;

    Abstract : Prostate cancer is the most common malignancy among Swedish men. Each year approx. 2 500 men die from the disease constituting almost 1/5 of all cancer death among men. Screening for prostate cancer has not been recommended in Sweden but PSA-testing is despite that commonly used. READ MORE
