  1. 1. Temporal Modulation in Fluorescence Spectroscopy and Imaging for Biological Applications

    Author : Gustav Persson; Jerker Widengren; Jörg Enderlein; KTH; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; fluorescence; spectroscopy; microscopy; modulated excitation; intensity modulation; fluorescence correlation spectroscopy; transient states; molecular kinetics; Physics; Fysik;

    Abstract : This thesis explores the benefits of intensity modulation for the purpose of extending the range of applications of fluorescence spectroscopy and imaging in cellular and molecular biology and medicine. Long-lived transient states of fluorescent molecules can, because of their long lifetimes, be used to detect subtle changes in the microenvironment of the molecule. READ MORE

  2. 2. Transient State Fluorescence Microscopy - method development and biological applications : Exploiting the dark states of fluorophores to measure oxygen concentrations, redox state, Förster resonance energy transfer and membrane viscosity

    Author : Thiemo Spielmann; Jerker Widengren; Karsten König; KTH; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Transient States imaging TRAST ; Triplet State imaging; fluorescence microscopy; modulated excitation; triplet state; radical state; trans-cis isomerisation;

    Abstract : Due to their long lifetime, triplet, redox and other transient states of fluorophores are highly sensitive to the micro-environment. Imaging their spatial distribution in biological samples can therefore help answer interesting questions about the metabolism, molecular interactions and dynamics in living cells. READ MORE

  3. 3. Monitoring Proton Exchange and Triplet States with Fluorescence

    Author : Tor Sandén; Jerker Widengren; Vogel Horst; KTH; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; fluorescence correlation spectroscopy; proton transfer; cytochrome c oxidase; transient state imaging; modulated excitation; Optics; Optik; Biochemistry; Biokemi;

    Abstract : Fluorescent molecules commonly shift to transient dark states, induced bylight or triggered by chemical reactions. The transient dark states can beused as probes of the local environment surrounding the fluorescent molecules,and are therefore attractive for use in biomolecular applications. READ MORE

  4. 4. Concepts and biomedical applications of excitation-modulated transient state monitoring of fluorescence emitters

    Author : Baris Demirbay; Jerker Widengren; Yves Mely; KTH; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Biological and Biomedical Physics; Biologisk och biomedicinsk fysik;

    Abstract : Fluorescence methods have developed very strongly in the last decade, allowing single- molecule detection sensitivity, high specificity, high time- and spatial resolution, as well as high readout speeds. Together, this makes fluorescencea very central readout modality for biomolecular and cellular studies. READ MORE

  5. 5. Time-Varying Excitation in Fluorescence Spectroscopy for Biological Applications

    Author : Gustav Persson; Jerker Widengren; Måns Ehrenberg; KTH; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; fluorescence; spectroscopy; molecular kinetics; Physics; Fysik;

    Abstract : The focus of this thesis is to explore and use the benefits of time-varying excitation in fluorescence spectroscopy for studies of biomolecular dynamics. Two new techniques taking advantage of modulated excitation are presented. READ MORE
