  1. 1. Neuronal and glial differentiation of expanded neural stem and progenitor cells; in vitro and after transplantation

    Author : Cecilia Eriksson Linsmeier; Neurobiologi; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Gtv-a; RCAS A -EGFP; transplantation; cell detection; Neurology; neuropsychology; neurophysiology; Neurologi; neuropsykologi; neurofysiologi; GFP-actin; glial differentiation; neuronal differentiation; neurospheres; glial cultures; bFGF; EGF; cell expansion; cortical primordium; MGE; LGE; CNS; stem- progenitor cells;

    Abstract : In this thesis we have used cells dissected from the lateral ganglionic eminence (LGE), the medial ganglionic eminence (MGE), and the cortical primordium of the embryonic mouse forebrain. The tissue was dissected from either i) wild-type mice, ii) green fluorescent protein (GFP)-, or iii) Gtv-a-expressing transgenic mice, and subsequently grown and expanded in vitro using two different protocols. 2. 2. Generation of midbrain dopaminergic neurons in vivo and in vitro: the role of Neurogenin2

    Author : Elin Andersson; Neurobiologi; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Medicine human and vertebrates ; Medicin människa och djur ; cell replacement; Parkinson?s; dopamine; neuronal specification; development; mesencephalon; TH; neuronal differentiation; proneural genes; transcription factors; bHLH; neurospheres; stem cell; progenitor cell;

    Abstract : Parkinsons disease (PD) is a neurodegenerative disorder where dopaminergic neurons of the substantia nigra (SNc) in the mesencephalon are progressively eliminated. The ensuing loss of dopaminergic innervation of the basal ganglia manifests itself as severe motor deficits in PD patients. READ MORE

  3. 3. Generation of cells for cell-replacement therapy: Specification of neural precursors in vivo and in neural stem cell cultures

    Author : Josephine Hebsgaard; Neurobiologi; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Neurospheres; Dopaminergic neurons; Parkinson s disease; Neurogenin2;

    Abstract : Cell replacement therapy of neurodegenerative disorders aims to substitute the degenerating cells with new functional neurons. Clinical trails with patients suffering from Parkinson’s or Huntington’s disease have generated proof-of-principle results that neural precursors taken from the developing human brain can survive upon grafting to the diseased brain and provide long-lasting symptomatic relief. 4. 4. Human neural stem cells: region-specific properties and prospects for cell therapy

    Author : Therese Kallur; Lund Neurologi; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Pax6; fetal cortex and striatum; human; neural stem cells; transplantation; neurospheres; stroke; overexpression; region-specific properties;

    Abstract : Cell replacement by neural transplantation can, in animal models of neurodegenerative diseases, reconstruct damaged brain circuitry. In the clinical situation, the graft material used for cell therapy must, most likely, be of human origin. READ MORE

  5. 5. Transcript profiling of small tissue samples using microarray technology

    Author : Maria Sievertzon; Joakim Lundeberg; Tom Freeman; KTH; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Biology; Gene expression analysis; transcriptomics; microarray analysis; 3’-tag signature amplification; neural stem cells; Biologi; Biology; Biologi;

    Abstract : Through a number of biological, technological and computational achievements during the 20th century and the devoted work of hundreds of researchers the sequence of the human and other genomes are now available in public databases. The current challenge is to begin to understand the information encoded by the DNA sequence, to elucidate the functions of the proteins and RNA molecules encoded by the genes as well as how they are regulated.
