  1. 1. Nucleic acid analysis tools : Novel technologies and biomedical applications

    Author : Iván Hernández-Neuta; Mats Nilsson; Frank Bier; Stockholms universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Nucleic acid analysis; padlock probes; selector probes; rolling circle amplification; microfluidics; single-molecule detection; digital quantification; fluorescence microscopy; mobile phone microscopy; molecular diagnostics; virology; Biochemistry; biokemi;

    Abstract : Nucleic acids are fundamental molecules of living organisms functioning essentially as the molecular information carriers of life. From how an organism is built to how it responds to external conditions, all of it, can be found in the form of nucleic acid sequences inside every single cell of every life form on earth. READ MORE

  2. 2. Structural and Biophysical Studies of Nucleic Acids

    Author : Wimal Pathmasiri; Adolf Gogoll; Lena Mäler; Uppsala universitet; []
    Keywords : Organic chemistry; nucleic acids; Nuclear Magnetic Resonance; molecular dynamics; sugar modified nucleosides; pKa; Organisk kemi;

    Abstract : This thesis is based on six research publications concerned with (i) study of the molecular structures and dynamics of modified nucleosides; (ii) investigation of the effect of incorporation of modified nucleosides on the structure of DNA; (iii) examination of the effect of the sugar modifications on the pseudo-aromatic properties (pKa) of the nucleobases; (iv) analysis of the effect of the CH-π interactions on the relative stability of the DNA-RNA hybrid duplexes. The structural stability of the nucleic acids as well as their behavior in molecular recognition is dominated by hydrogen bonding and stacking interactions beside other non-covalent interactions. 3. 3. Studies on Nucleic Acids – Structure and Dynamics

    Author : Johan Isaksson; Jyoti Chattopadhyaya; Douglas H. Turner; Uppsala universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Bioorganic chemistry; nucleic acids; modified nucleotides; NMR; structure; dangling-end; single-stranded; thermodynamics; hydration; Bioorganisk kemi; Bioorganic chemistry; Bioorganisk kemi;

    Abstract : This thesis is based on six papers, Papers I-VI, focusing on the interplay between the stabilizing elements of nucleic acids self-assembly; hydrogen bonding, stacking and solvent effects. In Paper I we investigate how the substitution of the O4' for CH2 in the sugar moiety of adenosine (2'-deoxyaristeromycin) at the A6 position of the Dickerson-Drew dodecamer makes the two modified bases exist in a dynamic equilibrium between Hoogsteen and Watson-Crick base pairing in the NMR time scale. 4. 4. Molecular Tools for Nucleic Acid Analysis

    Author : Deirdre O'Meara; KTH; []
    Keywords : nucleic acid capture; modular probes; biosensor; bio-magnetic separation; hepatitis C; sequencing; pyrosequencing; mutation detection; HIV-1; drug resistance; SNP; allele-specific extension; apyrase; genotyping.;

    Abstract : Nucleic acid technology has assumed an essential role invarious areas ofin vitrodiagnostics ranging from infectious diseasediagnosis to human genetics. An important requirement of suchmolecular methods is that they achieve high sensitivity andspecificity with a fast turnaround time in a cost-effectivemanner. READ MORE

  5. 5. Thermodynamics of Peptide Nucleic Acid Interactions with DNA

    Author : Tommi Ratilainen; Chalmers tekniska högskola; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; thermodynamics; enthalpy-entropy compensation; ionic strength; nucleic acids; fluorescence energy transfer; PNA; calorimetry; sequence-specific binding;

    Abstract : Peptide Nucleic Acid (PNA) is a DNA mimic with the natural nucleobases attached to a charge-neutral pseudopeptide backbone. It has remarkably strong and sequence-specific binding affinity to natural nucleic acids (DNA and RNA), mainly attributed to the lack of charge in the PNA backbone. READ MORE
