1. Thermodynamics of Peptide Nucleic Acid Interactions with DNA
Abstract : Peptide Nucleic Acid (PNA) is a DNA mimic with the natural nucleobases attached to a charge-neutral pseudopeptide backbone. It has remarkably strong and sequence-specific binding affinity to natural nucleic acids (DNA and RNA), mainly attributed to the lack of charge in the PNA backbone. READ MORE

2. Intelligent Nucleic Acid Interactions with Peptide Nucleic Acid (PNA) and Recombination Enzyme RecA
Abstract : The work presented in this thesis focuses on interactions between nucleic-acids in biological systems, particularly interactions of the promising drug-candidate peptide nucleic acid (PNA), and on protein-DNA intermediates during recombination. Several experimental techniques, mainly circular dichroism and fluorescence spectroscopy, have been used to characterize the interactions of interest. READ MORE

3. Acylated cell-penetrating peptides for nucleic acid delivery
Abstract : In recent decades many new methods have been developed to cure or treat genetical disorders such as cancer, viral infections or inheritable diseases. The problem is that the nucleic acids and their synthetic analogs, oligonucleotides, are not able to cross the cell membrane due to their physicochemical properties like high negative charge and size. 4. Characterization of nucleic acid delivery with fatty acid modified cell-penetrating peptide nanoparticle formulations
Abstract : Recent advances with techniques used for manipulating gene expression have brought us to an era where various gene therapeutic approaches are becoming common therapeutic tools for many previously incurable diseases. The main factor impeding the wider translation of gene therapy is that the active pharmaceutical ingredients used for interfering with gene expression are based on nucleic acids and synthetic oligonucleotides and such molecules do not readily reach their intracellular targets due to their physicochemical properties and therefore they require delivery vectors to cross the cell membrane. 5. A biophysical study of nucleic acid interactions with analogues and drugs
Abstract : work presented in this thesis concerns studies on the physicochemical nature of interactions between nucleic acids and small ligands. The outcome of such studies can yield insights at a molecular level into the physiological mechanisms of action of biologically active nucleic-acid binding molecules. READ MORE
