  1. 1. Confidence Predictions in Pharmaceutical Sciences

    Author : Staffan Arvidsson McShane; Ola Spjuth; Wesley Schaal; Lars Carlsson; Ernst Ahlberg; Stefan Kramer; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; machine learning; QSAR; Conformal prediction; Venn-ABERS; Probabilistic machine learning; Machine learning; Maskininlärning; Farmaceutisk vetenskap; Pharmaceutical Science;

    Abstract : The main focus of this thesis has been on Quantitative Structure Activity Relationship (QSAR) modeling using methods producing valid measures of uncertainty. The goal of QSAR is to prospectively predict the outcome from assays, such as ADMET (Absorption, Distribution, Metabolism, Excretion), toxicity and on- and off-target interactions, for novel compounds. 2. 2. Regulation of Vitamin D 25-hydroxylases : Effects of Vitamin D Metabolites and Pharmaceutical Compounds on the Bioactivation of Vitamin D

    Author : Maria Ellfolk; Kjell Wikvall; Maria Norlin; Olavi Pelkonen; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; CYP2D25; vitamin D 25-hydroxylase; transcriptional regulation; vitamin D; CYP2R1; CYP2J2; CYP27A1; phenobarbital; efavirenz; fibroblast; LNCaP; Pharmaceutical biochemistry; Farmaceutisk biokemi;

    Abstract : A 700bp portion of the promoter of CYP2D25, the porcine microsomal vitamin D 25-hydroxylase was isolated and sequenced. The computer analysis of the sequence revealed the existence of a putative VDRE at 220 bp upstream of the transcription start site. READ MORE

  3. 3. Muscarinic receptors and urinary bladder selectivity : Properties that determine the organ selectivity of compounds

    Author : Staffan Sundquist; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Pharmaceutical biosciences; bladder; muscarinic; receptor subtypes; smooth muscle; multivariate; Farmaceutisk biovetenskap; Biopharmacy; Biofarmaci; galenisk farmaci; Pharmaceutics;

    Abstract : Urinary bladder contraction is mainly mediated by activation of muscarinic acetylcholine receptors (mAChR). Muscarinic antagonists are often effective in treatment of unstable bladder. The effectiveness of this treatment has often been limited by side effects as dry mouth. READ MORE

  4. 4. Pharmaceutical binders and their function in directly compressed tablets : Mechanistic studies on the effect of dry binders on mechanical strength, pore structure and disintegration of tablets

    Author : Sofia Mattsson; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Pharmacy; Compaction; tablet; binder; pore structure; disintegration; FARMACI; PHARMACY; FARMACI; farmaceutisk farmakologi; Pharmaceutical Pharmacology;

    Abstract : In this thesis, the strength-enhancing mechanisms of dry binders in direct compression were studied. The systems investigated were binary mixtures containing various compounds and binders. Among the binders used were a series of different molecular weights of polyethylene glycol. READ MORE

  5. 5. Nasal administration of compounds active in the central nervous system : Exploring the olfactory system

    Author : Maria Dahlin; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Pharmacy; Nasal administration; olfactory pathway; cerebrospinal fluid; autoradiography; FARMACI; PHARMACY; FARMACI; farmaceutisk farmakologi; Pharmaceutical Pharmacology;

    Abstract : The nasal administration of drugs offers advantages over administration by intravenous injection. Drugs can be rapidly absorbed through the nasal mucosa, resulting in a rapid onset of action, and also avoiding degradation in the gastrointestinal tract and first-pass metabolism in the liver.
