  1. 1. The Interaction Between the Chaperonin GroEL and Human Carbonic Anhydrase II : Protein Folding and Stability Studies

    Author : Malin Persson; Linköpings universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; carbonic anhydrase; protein folding; protein stability; chaperone; GroEL; folding mechanism; site-directed spinlabeling; electron paramagnetic resonance EPR ;

    Abstract : The presence of GroEL/ES during the refolding of human carbonic anhydrase II was found to increase the yield of active enzyme from approx. 70 to approx. 100%. This chaperone action on the enzyme could be obtained by adding GroEL alone, and the time course in that case was only moderately slower than the spontaneous process. READ MORE

  2. 2. Perplexing Protein Puzzles

    Author : Stina Lindman; Biofysikalisk kemi; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; calbindin D9k; split-GFP; protein G B1; pKa-values; protein reconstitution; Electrostatic interactions; protein stability; protein denaturation; heteronuclear NMR; pH titrations;

    Abstract : Protein structure and stability are inherent in the amino acid sequence and governed by non-covalent interactions. The cooperation between forces is, however, perplexing and not well understood. In order to elucidate and predict protein folding and stability, detailed studies of non-covalent interactions are required. READ MORE

  3. 3. Structure determination and thermodynamic stabilization of an engineered protein-protein complex

    Author : Elisabet Wahlberg; Per-Åke Nygren; Bengt-Harald Jonsson; KTH; []
    Keywords : TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; affibody; protein structure; coupled folding; NMR spectroscopy; protein stability; protein-protein interactions; binding thermodynamics; isothermal titration calorimetry; protein engineering; Structural biochemistry; Strukturbiokemi;

    Abstract : The interaction between two 6 kDa proteins has been investigated. The studied complex of micromolar affinity (Kd) consists of the Z domain derived from staphylococcal protein A and the related protein ZSPA-1, belonging to a group of binding proteins denoted affibody molecules generated via combinatorial engineering of the Z domain. 4. 4. The birth and growth of the protein folding nucleus : Studies of protein folding focused on critical contacts, topology and ionic interactions

    Author : Linda Hedberg; Mikael Oliveberg; Christopher Johnson; Stockholms universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; protein folding; protein stability; two-state folding; chevron plot; transition-state movements; Hammond behaviour; topology; electrostatic interactions; pH dependence; mass-action; protein engineering; pKa; Biochemistry; Biokemi; biokemi; Biochemistry;

    Abstract : Proteins are among the most complex molecules in the cell and they play a major role in life itself. The complexity is not restricted to just structure and function, but also embraces the protein folding reaction. READ MORE

  5. 5. To Fold or To Fibrillate? Serendipity in Stability Studies

    Author : Olga Szczepankiewicz; Biofysikalisk kemi; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Protein stability; protein reconstitution; protein aggregation; Monellin; PGB1; Split-GFP; Amyloid beta;

    Abstract : Proteins are complex structures and years of research have been spent on attempts to understand their complexity. The non-covalent interactions involved in protein folding are: hydrophobic effect, electrostatic interactions, van der Waals interactions and hydrogen bonding. READ MORE
