1. Approaches to soft drug analogues of dihydrofolate reductase inhibitors : Design and synthesis
Abstract : The main objective of the research described in this thesis has been the design and synthesis of inhibitors of the enzyme dihydrofolate reductase (DHFR) intended for local administration and devoid of systemic side-effects. The blocking of the enzymatic activity of DHFR is a key element in the treatment of many diseases, including cancer, bacterial and protozoal infections, and also opportunistic infections associated with AIDS (Pneumocystis carinii pneumonia, PCP). 2. Selective Transformations of Allylic Compounds and Enol Derivatives : Synthesis of Fluorinated and Nitrogenated Scaffolds
Abstract : In this thesis, new synthetic methods to give access to molecules having fluorinated scaffolds and other polar functional groups have been developed. The methods give access to highly valuable organic compounds that may serve as building blocks in the synthesis of functionalized drug candidates. READ MORE

3. Palladium-promoted synthesis of compounds labelled with ¹¹C : Synthesis of ¹¹C-labelled prostacyclin and prostaglandin analogues
Abstract : Palladium-promoted reactions have been employed for the synthesis of compounds labelled with 11C (t½ = 20.3 min). The precursor [11C]methyl iodide was used in palladium-promoted cross-coupling reactions with organostannanes. READ MORE

4. Transformations of Nitrosoarenes and Alkynyl Enones : Selective Synthesis of Nitrogen-Containing Compounds
Abstract : The nitrogen atom plays a unique role in organic chemistry. It is abundantly found in organic materials and is responsible for the activity of many biologically relevant compounds. READ MORE

5. Diaryliodonium Salts : Development of Synthetic Methodologies and α-Arylation of Enolates
Abstract : This thesis describes novel reaction protocols for the synthesis of diaryliodonium salts and also provides an insight to the mechanism of α-arylation of carbonyl compounds with diaryliodonium salts. The first chapter gives a general introduction to the field of hypervalent iodine chemistry, mainly focusing on recent developments and applications of diaryliodonium salts. READ MORE
