  1. 1. Topoisomerase II and drug resistance in leukemic cells

    Author : Rong Zhou; Karolinska Institutet; Karolinska Institutet; []
    Keywords : ;

    Abstract : Mechanisms for resistance to chemotherapy include among other, elevated p-glycoprotein (p-gp) level and/or altered topoisomerase II level (topo II). The present study focused on topo 11 expression, and the enzyme related in vitro drug sensitivities in human myeloid leukemic K562 cell line and isolated peripheral mononuclear cells from patients with acute myelogenous leukemia (AML), and with chronic lymphocytic leukemia (CLL). 2. 2. Polyamines are involved in the regulation of S phase and DNA synthesis

    Author : Kersti Alm; Biologiska institutionen; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Djurfysiologi; Animal physiology; cell cycle regulation; p53; DNA strand breaks; replicon organization; topoisomerase II;

    Abstract : Our research group has previously shown that treatment with the polyamine biosynthesis inhibitors a-difluoromethylornithine (DFMO) or amidinoindan-1-one 2´-amidinohydrazone (CGP 48664) inhibited S phase progression before any other cell cycle phase was affected. This study was undertaken to further investigate the role of polyamines in the regulation of S phase progression and DNA synthesis. READ MORE

  3. 3. Chemosensitivity in Breast Cancer

    Author : Kenneth Villman; Carl Blomqvist; Ulf Tidefelt; Nils Wilking; Uppsala universitet; []
    Keywords : Oncology; breast cancer; chemotherapy; anthracycline; predictive; topoisomerase; TOP2A; HER2; cyclin A; in vitro; Onkologi;

    Abstract : Breast cancer mortality in Sweden is now in decline, thanks to early detection and the wide use of adjuvant endocrine therapy and chemotherapy. While hormone receptor status is predictive of response to endocrine treatment, there is no clinically useful predictive marker of a patient’s response to chemotherapy. READ MORE

  4. 4. Pharmacology and resistance mechanisms of nucleoside analogues and topoisomerase II interactive agents : studies on human leukemia cells with a focus on cross-resistance

    Author : Kourosh Lotfi; Rolf Larsson; Linköpings universitet; []
    Keywords : Nucleoside analogue; cross-resistance; multidrug resistance; deoxycytidine kinase; leukemia; anthracyclines; P-glycoprotein; MEDICINE; MEDICIN;

    Abstract : The purpose of this thesis was to elucidate mechanisms of action and resistance of clinically relevant nucleoside analogues and topoisomerase interactive agents in human leukemia cell lines and leukernia cells isolated from peripheral blood of leukemia patients. Interactions and cross-resistance patterns of these different cytotoxic drug families were also studied since these drugs are usually administrated in combination in the clinic. READ MORE

  5. 5. Investigating novel roles of topoisomerase regulatory interactions in transcription

    Author : Vladislav Kuzin; Karolinska Institutet; Karolinska Institutet; []
    Keywords : ;

    Abstract : Drugs targeting DNA Topoisomerases (TOPs, TOP1 and TOP2) are mainstays of anti-cancer therapy. While they have proven effective, the toxicity of current TOP drugs caused by DNA damage-induced apoptosis of non-cancer cells, limits their use in the clinic. READ MORE
