    Keywords : NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; NATURVETENSKAP; NATURAL SCIENCES; Thiopurine S-methyltransferase TPMT ; protein stability; enzyme kinetics; ligand binding; isothermal titration calorimetry ITC ; 8-anilinonaphthalene-1-sulfonic acid ANS ;

    Abstract : Thiopurine S-methyltransferase (TPMT) belongs to the Class I S-adenosylmethionine-dependent methyltransferase (SAM-MT) super family of structurally related proteins. Common to the members of this large protein family is the catalysis of methylation reactions using S-adenosylmethionine (SAM) as a methyl group donor, although SAM-MTs act on a wide range of different substrates and carry out numerous biologically important functions. 2. 2. Biophysical Characterization of Thiopurine S-Methyltransferase : A Key enzyme in the Effects of Thiopurine Drugs

    Author : Patricia Wennerstrand; Lars-Göran Mårtensson; Curt Peterson; Malin Lindqvist Appell; Mikael Widersten; Linköpings universitet; []
    Keywords : ;

    Abstract : In the treatment of leukemia and inflammatory bowel disease, thiopurines are commonly used drugs. Thiopurine S-methyltransferase (TPMT) is one of the drug metabolizing enzymes responsible of counteracting the formation of TGNs that will be incorporated into the DNA and RNA synthesis and thus induce apoptosis. 3. 3. Pharmacogenetic studies of thiopurines : focus on thiopurine methyltransferase

    Author : Malin Lindqvist Appell; Michel Eichelbaum; Linköpings universitet; []
    Keywords : MEDICINE; MEDICIN;

    Abstract : Pharmacogenetics represents the study ofvariability in drug response due to genetic variations. The thiopurines (6-mercaptopurine, 6-thioguanine and azathioprine) are prodrugs which require metabolic transformation to exert effect. READ MORE

  4. 4. Pharmacogenetic studies of thiopurine methyltransferase genotype-phenotype concordance and effect of methotrexate on thiopurine metabolism

    Author : Anna Zimdahl; Malin Lindqvist Appell; Lars-Göran Mårtensson; Inger Johansson; Linköpings universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES;

    Abstract : The thiopurines (6-mercaptopurine [6-MP], 6-thioguanine and azathioprine) are cytotoxic drugs used in the treatment of acute lymphoblastic leukemia (ALL), inflammatory bowel diseases, certain autoimmune diseases and after transplantation. The metabolism of thiopurines is complex with several enzymes involved in the conversion into active drug metabolites. READ MORE

  5. 5. Clinical studies of thiopurine metabolism in inflammatory bowel disease

    Author : Ulf Hindorf; Lund Medicin; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Gastroenterologi; Gastro-enterology; thiopurine methyltransferase; 6-mercaptopurine; azathioprine; Inflammatory bowel disease; thiopurines; adverse events; methylated metabolites; thioguanine nucleotides;

    Abstract : Inflammatory bowel disease (IBD, i.e. primarily Crohn's disease and ulcerative colitis) is characterised by a chronic or relapsing inflammation of the digestive tract. READ MORE
