  1. 1. Biophysical studies of cell-penetrating peptides and of the RNR inhibitor Sml1

    Author : Elsa Bárány-Wallje; Astrid Gräslund; Joachim Seelig; Stockholms universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; cell-penetrating peptides; transportan; bicelles; Sml1; Biophysics; Biofysik; Biophysics; biofysik;

    Abstract : Several short peptides, so called cell-penetrating peptides, have the capability to transport large hydrophilic cargos through the cell membrane. The objective is to use these peptides as drug carriers and thereby enhance the uptake of drugs into cells.Three different cell-penetrating peptides are characterized in this thesis. READ MORE

  2. 2. On cell-penetrating peptides and cell-junction interactions

    Author : Maria Lindgren; Nicholas Bodor; Stockholms universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; neurokemi och neurotoxikologi; Neurochemistry and Neurotoxicology;

    Abstract : Cell-penetrating peptides have the remarkable ability to carry hydrophilic macromolecules over the cellular membrane in an energy and protein independent manner. To synthesise and characterise new variants of cell-penetrating peptides, mainly aimed at finding analogues with minimised side effects and efficient cellular delivery, was one of the two main objectives of this thesis. 3. 3. The Application of isotropic bicelles as model membranes

    Author : August Andersson; Lena Mäler; G Arthur III Palmer; Stockholms universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Bicelle; membrane; peptide; spectroscopy; NMR; Molecular biophysics; Molekylär biofysik;

    Abstract : Isotropic bicelles are disc-shaped aggregates of lipids and detergents, and are suitable model systems for high-resolution NMR studies of membrane-interacting peptides. In this thesis the structures for the two peptides motilin and transportan were determined by homonuclear 1H methods in the presence of bicelles, and the structure of the bovine prion protein peptide (bPrPp) was solved in the presence of DHPC micelles. 4. 4. Cell-penetrating peptides and bioactive cargoes : Strategies and mechanisms

    Author : Kalle Kilk; Ülo Langel; Michael J. Gait; Stockholms universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Cell-penetrating peptide; delivery; Peptide nucleic acid; Biochemistry; Biokemi;

    Abstract : The cell membrane is an impermeable barrier for most macromolecules. Recently discovered cell-penetrating peptides (CPPs) have gained lot of attention because they can cross the membrane, and even more, carry cargoes with them. How CPPs enter cells is still not clear, while the delivery of different cargoes has been convincingly shown. READ MORE

  5. 5. Cell-penetrating peptides in model membrane systems : Interaction, structure induction and membrane effects

    Author : Mazin Magzoub; Astrid Gräslund; Steven Dowdy; Stockholms universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Molecular biophysics; Molekylär biofysik;

    Abstract : Despite continuing advances in the development of macromolecules, including peptides, proteins, and oligonucleotides, for therapeutic purposes, the successful application of these hydrophilic molecules has so far been hampered by their inability to efficiently traverse the plasma membrane. The discovery of a class of peptides (cell-penetrating peptides, CPPs) with the ability to mediate the non-invasive and efficient import of a whole host of cargoes, both in vitro and in vivo, has provided a new means by which the problem associated with cellular delivery can be circumvented.
