  1. 1. Towards the Development of Photoswitchable β-Hairpin Mimetics

    Author : Máté Erdélyi; Adolf Gogoll; Anders Karlén; John A. Robinson; Uppsala universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Organic chemistry; β-hairpin; peptidomimetics; Sonogashira; NMR; photochemistry; microwave; peptide; palladium; Organisk kemi; Organic chemistry; Organisk kemi;

    Abstract : Peptide secondary structure mimetics are important tools in medicinal chemistry, as they provide analogues of endogeneous peptides with new physicochemical and pharmacological properties. The β-hairpin motif has been shown to be involved in numerous physiological processes, among others in regulation of eucariotic gene transcription. READ MORE

  2. 2. The solution conformations of macrocycles. Applications in the exploration of weak interactions and in drug development

    Author : Emma Danelius; Göteborgs universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Macrocycles; cyclic peptides; NMR; solution conformational analysis; NAMFIS; β-hairpin; weak interactions; halogen bonding; protein-protein interaction; bioactive conformation; macrolides; cell permeable conformation.;

    Abstract : Understanding the solution conformation and dynamics of molecules with biological relevance, as well as the impact of their conformation stabilizing weak interactions, is for example important for drug design. Macrocycles have attractive pharmaceutical properties, and are of special interest as drug leads for targets with large, flat and featureless binding sites like protein-protein interfaces. READ MORE

  3. 3. Photoswitchable Peptidomimetics : Synthesis and Photomodulation of Functional Peptides

    Author : Miranda Varedian; Adolf Gogoll; Morten Grøtli; Uppsala universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; β-hairpin; peptidomimetics; stilbene; thioaurone; photochemistry; Organic chemistry; Organisk kemi;

    Abstract : The secondary structure of peptides is of pivotal importance for their biological function. The introduction of photoswitchable moieties into the backbones of peptides provides a unique way of regulating their conformation using an external stimulus, i.e., light. READ MORE

  4. 4. Inhibition of the MDM2/p53 Interaction Design, Synthesis and Evaluation of MDM2 Inhibitors

    Author : Mariell Pettersson; Göteborgs universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Protein-protein interactionProtein-protein interaction; MDM2 p53 interaction; MDM2 inhibitors; α- helix mimetics; β-hairpin; 2; 5-diketopiperazine; spiro-2; 5-diketopiperazine; purine; 8- triazolyl purine; solution conformational analysis; NAMFIS; fluorescence;

    Abstract : Numerous essential cellular processes are regulated by protein-protein interactions (PPIs) and PPIs have therefore been recognised as potential new drug targets. The transcription factor p53 is often referred to as the guardian of the genome due to its involvement in DNA repair, induction of cell cycle arrest and cellular apoptosis. READ MORE

  5. 5. Structural and biochemical basis for the high fidelity and processivity of DNA polymerase ε

    Author : Rais Ahmad Ganai; Erik Johansson; Per Elias; Umeå universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; DNA polymerase ε; Crystal structure; P domain; Pre-steady state kinetics; β-hairpin loop; Primer switching; Strand displacement; biokemi; Biochemistry;

    Abstract : DNA polymerase epsilon (Pol ε) is a multi-subunit B-family DNA polymerase that is involved in leading strand DNA replication in eukaryotes. DNA Pol ε in yeast consists of four subunits, Pol2, Dpb2, Dpb3, and Dpb4. Pol2 is the catalytic subunit and Dpb2, Dpb3, and Dpb4 are the accessory subunits. READ MORE
