  1. 1. Disease-causing Keratin Mutations and Cytoskeletal Dysfunction in Human Skin : In vitro Models and new Pharmacologic Strategies for Treating Epidermolytic Genodermatoses

    Author : Jean Christopher Chamcheu; Anders Vahlquist; Hans Törmä; Harshad Navsaria; Marie Virtanen; Torbjörn Egelrud; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; epidermolysis bullosa simplex; epidermolytic ichthyosis; genodermatoses; keratin; keratin mutation; keratinocytes; gene therapy; pharmacological therapy; immortalization; gene regulation; trimethylamine N-oxide TMAO ; sodium 4-phenylbutyrate 4-PBA ; tissue engineering; cell culture; heat shock proteins; MAP kinases; Dermatology and venerology; Dermatologi och venerologi; Dermatologi och venereologi; Dermatology and Venerology;

    Abstract : Epidermolysis bullosa simplex (EBS) and epidermolytic ichthyosis (EI) are rare skin fragility diseases characterized by intra-epidermal blistering due to autosomal dominant-negative mutations in basal (KRT5 or KRT14) and suprabasal (KRT1 or KRT10) keratin genes,  respectively. Despite vast knowledge in the disease pathogenesis, the pathomechanisms are not fully understood, and no effective remedies exist. READ MORE

  2. 2. Neural stem cell-mediated enhanced gene therapy of malignant gliomas using a novel plant thymidine kinase

    Author : Zahidul Khan; Karolinska Institutet; Karolinska Institutet; []
    Keywords : Malignant glioma; suicide gene therapy; bystander effect; gap junction; neural stem cell; 4-phenylbutyrate; epigenetics; cell differentiation;

    Abstract : Malignant gliomas are the most common brain tumors in adults. Approximately, 190,000 new patients are diagnosed with malignant gliomas worldwide every year. Unfortunately, the prognosis of malignant gliomas still remains very poor, despite advances in management. READ MORE

  3. 3. Experimental combination therapy of brain cancer cell models

    Author : Ana-Maria Marino; Karolinska Institutet; Karolinska Institutet; []
    Keywords : ;

    Abstract : Epigenetic alterations and aberrant expression of genes controlling epigenetic mechanisms have been identified in several cancers, including medulloblastoma and glioma, the most common primary brain tumors in children and adults, respectively. We have been investigating if combination therapy using histone deacetylase inhibitors (HDACi) and receptor tyrosine kinase inhibitors (RTKi) will enhance glioblastoma and medulloblastoma cell killing. 4. 4. Two novel combination therapies of glioblastoma multiforme from a Cx43 perspective : studies of in vitro models

    Author : Thomas Asklund; Karolinska Institutet; Karolinska Institutet; []
    Keywords : Glioma; gap junction; Cx43; bystander effect; HSV-tk; HDAC inhibitor; tyrosine kinase inhibitor;

    Abstract : The group of patients suffering from the malignant brain tumour glioblastoma multiforme (GBM) have a median survival of only about one year. Thus, new treatment strategies are urgently needed. Gap junctions are intercellular channels, permitting intercellular transfer of water soluble substances. READ MORE

  5. 5. Nephrin cellular trafficking and intracellular interactions

    Author : Xiao Li Liu; Karolinska Institutet; Karolinska Institutet; []
    Keywords : Nephrin; signal; missense mutations; trafficking.;

    Abstract : Nephrin is a 180-kDa transmembrane glycoprotein belonging to the immunoglobulin family of cell adhesion molecules. Nephrin has an extracellular part with eight Ig-like motifs, one fibronectin-like domain, a transmembrane region and a short cytoplasmic tail. READ MORE
