  1. 1. Pharmacogenetic studies of thiopurine methyltransferase genotype-phenotype concordance and effect of methotrexate on thiopurine metabolism

    Author : Anna Zimdahl; Malin Lindqvist Appell; Lars-Göran Mårtensson; Inger Johansson; Linköpings universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES;

    Abstract : The thiopurines (6-mercaptopurine [6-MP], 6-thioguanine and azathioprine) are cytotoxic drugs used in the treatment of acute lymphoblastic leukemia (ALL), inflammatory bowel diseases, certain autoimmune diseases and after transplantation. The metabolism of thiopurines is complex with several enzymes involved in the conversion into active drug metabolites. READ MORE

  2. 2. Pharmacological and molecular investigations on the mechanisms underlying resistance of human leukaemia cells to the antimetabolites methotrexate, 6-mercaptopurine and 6-thioguanine

    Author : Alan Kambiz Fotoohi; Karolinska Institutet; Karolinska Institutet; []
    Keywords : ;

    Abstract : The goal of this thesis has been to elucidate the mechanisms underlying resistance to methotrexate (MTX), 6-mercaptopurine (6-MP) and 6-thioguanine (6-TG), the antimetabolites widely used in treatment of childhood leukemia, as well as to determine the influence of 7-hydroxymethotrexate (7-OHMTX), the major metabolite of MTX, on the therapeutic action of MTX. Resistant sublines of leukemic cell lines were developed by long-term exposure to stepwise increasing concentrations of these different agents. READ MORE

  3. 3. Etiology and risk factors of febrile neutropenia in children during cancer treatment

    Author : Martina Wahlund; Karolinska Institutet; Karolinska Institutet; []
    Keywords : ;

    Abstract : Side effects due to chemotherapy is still a major issue during cancer treatment. Febrile neutropenia and associated microbiological defined infections (MDIs) are dreaded complications, and still a cause of death during pediatric cancer treatment. READ MORE

  4. 4. Pharmacological studies on uptake, metabolism, and resistance to anti-cancer drugs : insights into the treatment of leukemia

    Author : Hazhar Karim; Karolinska Institutet; Karolinska Institutet; []
    Keywords : ;

    Abstract : Leukemia is blood cancer that begins in the bone marrow and characterized by abnormal production of white blood cells. Due to advances in treatment, there has been a dramatic increase in survival rate for patients with different types of leukemia. READ MORE

  5. 5. Interindividual differences in thiopurine metabolism : studies with focus on inflammatory bowel disease

    Author : Sofie Haglund; Sven Almer; Jan Söderman; Curt Peterson; Åke Nilsson; Linköpings universitet; []
    Keywords : MEDICINE; MEDICIN;

    Abstract : The thiopurines, 6-mercaptopurine and its prodrug azathioprine, are used in the treatment of inflammatory bowel disease, ulcerative colitis and Crohn´s disease. The main active metabolites are the phosphorylated thioguanine nucleotides (6-TGNs) and methylated thioinosine monophosphate (meTIMP). READ MORE
