  2. 2. Interplay between Transcription and Homologous Recombination in the Presence of DNA Damage

    Author : Ivaylo Stoimenov; Thomas Helleday; Niklas Schultz; Natalia Issaeva; Thierry Nouspikel; Stockholms universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; homologous recombination; transcription; DRB; UVC; 6-thioguanine; mammalian cells; BRCA1; BRCA2; PARP inhibitors; XP-V; Molecular biology; Molekylärbiologi; molekylärgenetik; Molecular Genetics;

    Abstract : The biochemical processes of DNA repair, replication and recombination compete for the same substrate, the DNA molecule. This competition is natural, as each process requires the same template. In order to resolve possible conflicts between these processes, when they take place on a particular stretch of DNA, certain crosstalk is expected. READ MORE

  3. 3. Pharmacological and molecular investigations on the mechanisms underlying resistance of human leukaemia cells to the antimetabolites methotrexate, 6-mercaptopurine and 6-thioguanine

    Author : Alan Kambiz Fotoohi; Karolinska Institutet; Karolinska Institutet; []
    Keywords : ;

    Abstract : The goal of this thesis has been to elucidate the mechanisms underlying resistance to methotrexate (MTX), 6-mercaptopurine (6-MP) and 6-thioguanine (6-TG), the antimetabolites widely used in treatment of childhood leukemia, as well as to determine the influence of 7-hydroxymethotrexate (7-OHMTX), the major metabolite of MTX, on the therapeutic action of MTX. Resistant sublines of leukemic cell lines were developed by long-term exposure to stepwise increasing concentrations of these different agents. READ MORE

  4. 4. Optimizing Chemotherapy in Childhood Acute Myeloid Leukemia

    Author : Josefine Palle; Gudmar Lönnerholm; Britt-Marie Frost; Rolf Larsson; Henrik Schröder; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; childhood; acute myeloid leukemia; drug resistance; pharmacokinetics; chromosomal abnormalities; Down syndrome; MLL-rearrangement; t 9; 11 ; Paediatric medicine; Pediatrisk medicin;

    Abstract : Despite major advances in our understanding of the biology of childhood acute myeloid leukemia (AML) and the development of new cytotoxic drugs, the prognosis of long-term survival is still only 60-65 %.In the present research, we studied the pharmacokinetics of drugs used in the induction therapy of childhood AML and performed in vitro drug sensitivity testing of leukemic cells from children with AML. 5. 5. Pathogenic mechanisms affecting mitochondrial DNA replication and transcription

    Author : Meenakshi Singh; Göteborgs universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; mtDNA; ribonucleotides; nucleoside analogues; POLRMT;

    Abstract : Mitochondria are cytoplasmic organelles fundamental to life and health. In mitochondria, energy from the food we eat is converted into adenosine triphosphate (ATP), which in turn is used as a source of chemical energy to drive a multitude of cellular reactions. READ MORE
