  1. 1. Studies on mutagenesis by ethylene oxide and styrene oxide in the hprt gene in human cells

    Author : Tatiana Bastlova; Karolinska Institutet; Karolinska Institutet; []
    Keywords : ethylene oxide; styrene; styrene-7; 8-oxide; human diploid fibroblasts; human T-lymphocytes; hprt mutation;

    Abstract : Environmental, occupational and life style-related exposures to mutagenic agents may contribute to cancer risk in humans. To prevent the potentially hazardous effects of such agents it is important to understand their mechanisms of action. READ MORE

  2. 2. Evaluation of toxic and genotoxic effects of xenobiotica by means of a liver perfusion system

    Author : Brita Beije; Stockholms universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES;

    Abstract : An isolated liver perfusion system is described in which mammalian cells, as genetic targets, can be directly and continuously exposed to the perfusate at varying distances from the metabolising organ. Thus, potentially mutagenic metabolites released from the liver can be detected, and the biological half-life of the reactive metabolites can be estimated. READ MORE

  3. 3. Evolutionary Analysis and Posttranslational Chemical Modifications in Protein Redesign : A Study on Mu Class Glutathione Transferases

    Author : Ylva Ivarsson; Bengt Mannervik; Gun Stenberg; Richard N. Armstrong; Uppsala universitet; []
    Keywords : Biochemistry; glutathione transferase; evolutionary analysis; positive selection; epoxide; stereoselectivity; protein modification; Biokemi;

    Abstract : Glutathione transferases (GSTs) constitute a family of multifarious enzymes that conjugate glutathione (GSH) with a wide range of electrophiles. GSTs are grouped into different classes based on protein sequence similarities. Despite high sequence identities between GSTs of the same class they often display different substrate specificites. 4. 4. Microsomal epoxide hydrolase : characterization, induction, and in vitro modulation, of the catalytic activity

    Author : Janeric Seidegård; Stockholms universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES;

    Abstract : Microsomal epoxide hydrolase has been characterized, mainly in the fiver and lung of the rat.A reported radiometric assay using 3H-styrene 7,8-oxide as substrate has been modified by purifying the substrate before use, decreasing the incubation volume, and prolonging the incubation time. 5. 5. Studies on hepatic glutathione transferases from rat and man : 1. A kinetic mechanism for glutathione transferase A from rat liver. 2. Transferase µ, a new form of glutathione transferase in human liver

    Author : Margareta Warholm; Stockholms universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES;

    Abstract : This study of glutathione transferases was undertaken to achieve a better understanding of the catalytic and molecular properties of various isozymes of this group of drug-metabolizing enzymes.The steady-state kinetics of purified glutathione transferase A from rat liver with glutathione (GSH) and 1,2-dichloro- 4-nitrobenzene as substrates, did not follow a simple Michaelis- Menten equation.
