  1. 1. Aldehyde toxicity in human oral epithelium

    Author : Jan Anders Nilsson; Karolinska Institutet; Karolinska Institutet; []
    Keywords : Oral epithelium; cell cultures; toxicity; transformation; aldehydes; thiols; ADH3;

    Abstract : Human oral epithelium is an established target for aldehyde toxicity, including from usage of tobacco, intake of foods and fluids, leaching of dental materials, and from mouth inhalation of aldehydes in the work environment. Following establishment of culture/exposure protocols for oral cell cultures, toxicity mechanisms and risk of malignant transformation from aldehyde exposure was investigated, including the metabolic defense offered by alcohol dehydrogenase 3 (ADH3) against formaldehyde toxicity. 2. 2. Function, expression and polymorphism of human alcohol dehydrogenase 3/glutathionedependent formaldehyde dehydrogenase

    Author : Jesper Hedberg; Karolinska Institutet; Karolinska Institutet; []
    Keywords : ;

    Abstract : Alcohol dehydrogenase 3 (ADH3), identical to glutathione-dependent formaldehyde dehydrogenase, plays a vital role in the defense against formaldehyde through the activity for the spontaneously formed adduct between formaldehyde and glutathione, Shydroxymethylglutathione (HMGSH). ADH3 may also participate in the metabolism of Snitrosylated glutathione, GSNO. READ MORE

  3. 3. Gene expression in buccal keratinocytes with emphasis on carbonyl metabolism

    Author : Claudia A Staab; Karolinska Institutet; Karolinska Institutet; []
    Keywords : ;

    Abstract : The inner lining of the cheek, the buccal mucosa, is a target for air-borne, dietary and tobacco usage-derived carcinogens, but also interesting from a drug delivery point of view. Cancer arising in the buccal epithelium, buccal squamous cell carcinoma (BSCC), often diagnosed at a late disease stage, is highly aggressive and recurrent, emphasizing the need for novel approaches in diagnosis and therapy. 4. 4. Medium chain dehydrogenases/reductases : alcohol dehydrogenases of novel types

    Author : Annika Norin; Karolinska Institutet; Karolinska Institutet; []
    Keywords : Alcohol dehydrogenase; Nicotinoproteins; Structural interpretations; Functional properties; Molecular evolution.;

    Abstract : The medium chain dehydrogenase/reductase (MDR) superfamily is separated into at least eight families, of which the dimeric alcohol dehydrogenase (ADH) is one. Including species variants, close to 1000 MDR forms have been characterized. READ MORE

  5. 5. Enzymatic studies of alcohol dehydrogenase by a combination of in vitro and in silico methods

    Author : Mikko Hellgren; Karolinska Institutet; Karolinska Institutet; Sweden Stockholm Karolinska Institutet Department of Medical Biochemistry and Biophysics; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES;

    Abstract : The family of alcohol dehydrogenases (ADHs) catalyzes conversions of alcohols, ketons and aldehydes. The early discovery and isolation of ADH (1937) was followed by numerous investigations. It was also the first dimeric enzyme for which the threedimensional structure was determined (1974). READ MORE
