  1. 1. Immune reconstitution after childhood leukemia. Aspects on immunizations and effects of Ara-C on the innate immune system

    Author : Torben Ek; Göteborgs universitet; []
    Keywords : acute lymphoblastic leukemia; immune reconstitution; immunization; antibody; avidity; ara-C; fever; cytokines;

    Abstract : Children with acute lymphoblastic leukemia (ALL) can be cured with cytotoxic chemotherapy, but myelosuppression and immunosuppression are major side effects causing morbidity and even mortality from infections. Vaccinations with diphtheria toxoid (DT), tetanus toxoid (TT) and protein conjugated Haemophilus influenzae type B (Hib) capsular polysaccharide were used to investigate the adaptive immune system in a controlled study of 31 children after treatment for ALL. 2. 2. Pharmacological and clinical studies of new ways to improve cytostatic treatment of acute myelocytic leukemia : in vitro and in vivo studies

    Author : Christina Löfgren; Karolinska Institutet; Karolinska Institutet; []
    Keywords : ;

    Abstract : Treatment failure in refractory or relapsed disease is often caused by resistance towards Ara-C or daunorubicin, the two drugs mainly used in the treatment of acute myeloid leukemia (AML). This thesis comprises studies of resistance mechanisms regarding Ara-C and the newer nucleoside analogues CdA and fludarabine, improvement of treatment outcome after enhancement of tumour AraC susceptibility by concomitant treatment with GM-CSF and the effect of liposomal formulation on leukemic cell uptake of daunorubicin. 3. 3. In vitro studies of cytostatic drugs and growth factors on leukemic and normal hematopoietic cells : based on in vivo intracellular pharmacokinetics

    Author : Britt Sundman-Engberg; Karolinska Institutet; Karolinska Institutet; []
    Keywords : ANLL; Normal bone marrow; in vitro; predictive tests; pharmacokinetics; cytostatic drugs; growthfactors;

    Abstract : The aims were to develop, evaluate and apply in vitro methods for cytostatic drug sensitivity, based on intracellular concentrations found after in vivo infusions. The intracellular pharmacokinetics of anthraquinones and ara-C were compared during and after infusion to leukemic patients. Drug concentrations were evaluated with HPLC. READ MORE

  4. 4. Identification and characterization of novel genetic and epigenetic factors required for normal and malignant hematopoiesis

    Author : Yaser Heshmati; Karolinska Institutet; Karolinska Institutet; []
    Keywords : ;

    Abstract : Acute myeloid leukemia (AML) is a type of blood cancer, characterized by clonal expansion and loss of differentiation ability of myeloid progenitor cells leading to abnormal accumulation of immature myeloid cells (myeloblasts) in the bone marrow and peripheral blood. This thesis (study I to IV) focused on the identification and characterization of genes which are required for AML growth. READ MORE

  5. 5. On mechanisms of drug resistance in acute myeloid leukemia

    Author : Malin Prenkert; Ulf Tidefelt; Hilja Strid; Mats Merup; Örebro universitet; []
    Keywords : MEDICINE; MEDICIN; Biomedicin; Biomedicine;

    Abstract : In this thesis focus has been to increase the knowledge and understanding of some of the mechanisms responsible for drug resistance in acute myeloid leukemia, as well as identify possibilities to predict drug resistance at diagnosis.We have studied the intracellular behavior of cytostatic drugs and their main metabolites (paper I) and the cellular response to cytostatic drugs (paper III).
