  1. 1. Amyloid-β Protofibril Formation and Neurotoxicity : Implications for Alzheimer’s Disease

    Author : Ann-Sofi Johansson; Lars Lannfelt; Frida Ekholm Pettersson; Pär Gellerfors; Tobias Hartmann; Uppsala universitet; []
    Keywords : Neurosciences; Amyloid-β; Neurotoxicity; Aggregation; Protofibrils; Alzheimer s disease; Neurovetenskap;

    Abstract : Alzheimer’s disease (AD) is the most common cause of dementia. A characteristic feature of AD is the presence of amyloid plaques in the cortex and hippocampus of the brain. The principal component of these plaques is the amyloid-β (Aβ) peptide, a cleavage product from proteolytic processing of amyloid precursor protein (APP). READ MORE

  2. 2. Capturing transient peptide assemblies associated with Alzheimer's disease : Native mass spectrometry studies of amyloid-β oligomerization

    Author : Nicklas Österlund; Astrid Gräslund; Leopold L. Ilag; Mikael Oliveberg; Sheena E. Radford; Stockholms universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; Protein aggregation; amyloid-β; neurodegeneration; Alzheimer’s disease; chaperones; DnaJ; biomembranes; native mass spectrometry; Biophysics; biofysik;

    Abstract : Correct folding of proteins is essential for maintaining a functional living cell. Misfolding and aggregation of proteins, where non-native intermolecular interactions form large and highly ordered amyloid aggregates with low free energy, is hence associated with multiple diseases. 3. 3. Modeling Amyloid-β Pathology in Alzheimer’s Disease Using the Arctic Mutation

    Author : Ola Philipson; Lars Nilsson; Lars Lannfelt; Fred van Leuven; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Alzheimer’s disease; Amyloid; Amyloid-β; intraneuronal; transgenic mice; immunohistochemistry; ELISA; Public health medicine research areas; Folkhälsomedicinska forskningsområden; Geriatrics; Geriatrik;

    Abstract : The Arctic mutation in the Amyloid-β (Aβ) domain of the Amyloid-β precursor protein (APP) causes Alzheimer’s disease (AD) and confers unique biochemical characteristics to Aβ peptides. The aims of this thesis were to evaluate a transgenic model with the Arctic mutation, and to use it to gain new insights into the mechanisms of early (pre-plaque) and late-stage Aβ pathogenesis in AD. 4. 4. Investigation of the intercellular transmission of α-synuclein, amyloid-β and TDP-43

    Author : Christopher Sackmann; Martin Hallbeck; Emelie Severinsson; Poul Henning Jensen; Linköpings universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; α-synuclein; amyloid-β; TDP-43; cell to cell transmission;

    Abstract : Neurodegenerative diseases such as Alzheimer’s disease (AD), Parkinson’s disease (PD), frontotemporal lobar dementia (FTLD) and amyotrophic lateral sclerosis (ALS) are disorders characterized by the progressive deposition of proteinaceous inclusions throughout the brain in a predictable manner. Each disease is described by the involvement of different misfolded and aggregated proteins (AD, amyloid-β and tau; PD, α-synuclein; ALS and FTLD, TDP-43) that spread between anatomically connected brain regions, causing cell death in previously healthy regions. 5. 5. On the molecular mechanisms of the amyloid β-peptide aggregation

    Author : Kalyani Sanagavarapu; NanoLund: Centre for Nanoscience; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; protein aggregation; amyloid β-peptide; aggregation kinetics; aggregation mechanism N-terminus; mutations; variants; sequence determinants; ; cross-seeding;

    Abstract : The pathogenesis of Alzheimer’s disease is widely believed to be due to production and deposition of the amyloid β-peptide. Several variants of the Aβ peptide are known to exist in in vivo. Variations include mutations or additional functional groups attached to residue side chains and may affect the aggregation process.
