  1. 1. Islet amyloid polypeptide (IAPP) in Type 2 diabetes and Alzheimer disease

    Author : Marie Oskarsson; Gunilla Westermark; Mikael Oliveberg; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; amyloid; IAPP; Abeta; type 2 diabetes; Alzheimer disease; BRICHOS; heparan sulfate; islet amyloid; amyloid plaques; seeding; Medical Cell Biology; Medicinsk cellbiologi;

    Abstract : The misfolding and aggregation of the beta cell hormone islet amyloid polypeptide (IAPP) into amyloid fibrils is the main pathological finding in islets of Langerhans in type 2 diabetes. Pathological assemblies of IAPP are cytotoxic and believed to contribute to the loss of insulin-producing beta cells. READ MORE

  2. 2. Of spiders, bugs, and men : Structural and functional studies of proteins involved in assembly

    Author : Wangshu Jiang; Stefan D Knight; Steve J. Matthews; Uppsala universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; Protein assembly; sulfate transporter; crystallography; Proteus mirabilis; fimbriae; adhesin; urinary tract infection; biofilm; spider silk; asymmetric dimer; Bri2; BRICHOS; molecular chaperone; Alzheimer s disease; amyloid; Biology with specialization in Structural Biology; Biologi med inriktning mot strukturbiologi;

    Abstract : Protein assembly enables complex machineries while being economical with genetic information. However, protein assembly also constitutes a potential threat to the host, and needs to be carefully regulated.Sulfate is a common source of sulfur for cysteine synthesis in bacteria. READ MORE

  3. 3. Advances in Ligand Binding Predictions using Molecular Dynamics Simulations

    Author : Henrik Keränen; Johan Åqvist; Jordi Villà i Freixa; Uppsala universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; free-energy perturbation; molecular dynamics; ligand binding; free-energy perturbation; linear interaction energy; binding free-energy; homology modeling; virtual screening; alanine scanning; amino acid mutagenesis; hERG; GPCR; adenosine receptor; serotonin receptor; BRICHOS; cruzain;

    Abstract : Biochemical processes all involve associations and dissociations of chemical entities. Understanding these is of substantial importance for many modern pharmaceutical applications. In this thesis, longstanding problems with regard to ligand binding are treated with computational methods, applied to proteins of key pharmaceutical importance. READ MORE

  4. 4. Islet Amyloid Polypeptide: Interaction with Amyloid Beta, Alpha-Synuclein and BRICHOS

    Author : Ye Wang; Gunilla Westermark; Ludmilla Morozova-Roche; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; amyloid; IAPP; Abeta; alpha-synuclein; Bri2 BRICHOS; luminescent conjugated oligothiophenes; Medical Cell Biology; Medicinsk cellbiologi;

    Abstract :  Amyloid, the congophilic deposits of misfolded protein, are pathological hallmarks of many common diseases, among others, Alzheimer’s disease (AD), Parkinson’s disease (PD), and type 2 diabetes (T2D). Amyloid beta (Aβ) forms senile plaques in AD, alpha-synuclein (aSyn) forms Lewy bodies and Lewy neurites in PD, islet amyloid polypeptide (IAPP) forms islet amyloid in T2D. READ MORE

  5. 5. BRICHOS interactions with amyloid proteins and implications for Alzheimer disease

    Author : Lisa Dolfe; Karolinska Institutet; Karolinska Institutet; []
    Keywords : ;

    Abstract : To date, about 30 diseases, in which amyloid fibrils form extracellular deposits, have been identified in humans. It is not known if the fibrils have a function, like storage of misfolded proteins, or if they just reflect failure of the cell to manage misfolded proteins. READ MORE
