  1. 1. Developmental Aspects of Drug Transport Across the Blood-Brain Barrier

    Author : Jörgen Bengtsson; Margareta Hammarlund-Udenaes; Ann-Cathrin Engwall; Sven Björkman; Elizabeth C.M. de Lange; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Blood-brain barrier; development; active transport; tight junction proteins; microdialysis; recovery; morphine; nitrofurantoin; Bcrp; microarray; real-time PCR; in vitro; in vivo; LC-MS MS; Biopharmacy; Biofarmaci; Pharmacokinetics and Drug Therapy; Farmakokinetik och läkemedelsterapi;

    Abstract : The developmental aspect of drug transport across the blood-brain barrier (BBB) was investigated. Microdialysis was used to study unbound morphine BBB transport at different ages in sheep. An in vitro study was performed to find differentially expressed genes in brain capillary-rich fractions of the brain in rats of different ages. READ MORE

  2. 2. ATP-Binding Cassette Efflux Transporters and Passive Membrane Permeability in Drug Absorption and Disposition

    Author : Pär Matsson; Per Artursson; Ulf Norinder; Peter Swaan; Uppsala universitet; []
    Keywords : Pharmaceutics; ATP-binding cassette; ABC transporter; P-gp; P-glycoprotein; ABCB1; BCRP; Breast cancer resistance protein; ABCG2; MRP2; Multidrug-resistance associated protein 2; ABCC2; Membrane permeability; Drug transport; Active transport; Passive diffusion; Multivariate data analysis; PLS; OPLS; QSAR; Galenisk farmaci;

    Abstract : Transport into and across the cells of the human body is a prerequisite for the pharmacological action of drugs. Passive membrane permeability and active transport mechanisms are major determinants of the intestinal absorption of drugs, as well as of the distribution to target tissues and the subsequent metabolism and excretion from the body. 3. 3. ATP-Binding-Cassette Transporters in Biliary Efflux and Drug-Induced Liver Injury

    Author : Jenny M. Pedersen; Per Artursson; Christel A. S. Bergström; Stefan Oswald; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; ABC transport protein; Pgp; P-glycoprotein; ABCB1; BCRP; breast cancer resistance protein; ABCG2; MRP2; multidrug resistance-associated protein 2; ABCC2; BSEP; bile salt export pump; ABCB11; sandwich cultured human hepatocytes; SCHH; drug-induced liver injury; DILI; multivariate data analysis; OPLS; Galenisk farmaci; Pharmaceutics;

    Abstract : Membrane transport proteins are known to influence the absorption, distribution, metabolism, excretion and toxicity (ADMET) of drugs. At the onset of this thesis work, only a few structure-activity models, in general describing P-glycoprotein (Pgp/ABCB1) interactions, were developed using small datasets with little structural diversity. 5. 5. Interindividual Variability of Drug Transport Proteins : Focus on Intestinal Pgp (ABCB1) and BCRP (ABCG2)

    Author : Gunilla Englund; Per Artursson; Håkan Melhus; Andreas Kindmark; Joseph W Polli; Uppsala universitet; []

    Abstract : The appearance of adverse drug reactions is a common reason for hospitalization in Western countries. Research on underlying biological mechanisms for interindividual variability in drug response aims to better identify patients with exceptional genetic traits, disease conditions or risk of drug-drug interactions and thereby help to prevent adverse drug reactions.
