  1. 1. The Plant Plasma Memrbane H+-ATPase: regulation by phosphorylation and 14-3-3 proteins

    Author : Anne Olsson; Biokemi och Strukturbiologi; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; phosphorylation; fusicoccin; 14-3-3; C-terminus; autoinhibitory domain; plasma membrane; plant; H -ATPase; Plant biochemistry; Växtbiokemi;

    Abstract : The plant plasma membrane H+-ATPase is a predominant membrane enzyme that provides the energy for secondary active transport across the plasma membrane. Consequently, the H+-ATPase is thought to play a major role in many cell processes, and it is implicated to be regulated by a number of factors, including hormones, blue light, and fungal toxins The plant plasma membrane H+-ATPase is regulated via an autoinhibitory domain located within the C-terminal region of the enzyme. 2. 2. Nucleotide Binding to Trimeric dUTPase: Studies on Recombinant Enzymes using Kinetic and Spectroscopic Methods

    Author : Johan Nord; Biokemi och Strukturbiologi; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; C-terminus; dUTPase; EIAV; Escherichia coli; flexibility; Homo sapiens; kinetics; NMR; nucleotide metabolism; rate constants of lig-and binding; production of recombinant enzyme; dUTP; specificity; Biochemistry; Metabolism; Biokemi;

    Abstract : Recombinant dUTPase from the bacterium Escherichia coli, the retrovirus equine infectious anemia virus (EIAV) and Homo sapiens have been made available for physico-chemical characterisation. The protocols developed have also proven to be suitable for production of enzymes modified by isotope labelling or other methods. READ MORE

  3. 3. The Plasma Membrane H+-ATPase - Identification of a 14-3-3 binding motif

    Author : Fredrik Svennelid; Biokemi och Strukturbiologi; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; regulation; Plant biochemistry; Växtbiokemi; plasma membrane; plant; phosphorylation; isoforms; H -ATPase; fusicoccin; C terminus; binding motif; 14-3-3; autoinhibitory domain;

    Abstract : The P-type plasma membrane H+-ATPases form a group of proteins only found in plants and fungi. The pumping of protons across the plasma membrane, energized by ATP hydrolysis, creates an electrochemical gradient that is essential for solute transport and internal pH regulation. READ MORE

  4. 4. Redesign of Alpha Class Glutathione Transferases to Study Their Catalytic Properties

    Author : Lisa O Nilsson; Ralf Morgenstern; Uppsala universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Biochemistry; glutathione transferase; heterologous expression; heterodimer; hydroxyalkenal; protein redesign; sequential design; dimer-interface interactions; enzyme kinetics; glutathione; dynamic C-terminus; viscosity dependence; Biokemi; Biochemistry; Biokemi; biokemi; Biochemistry;

    Abstract : A number of active site mutants of human Alpha class glutathione transferase A1-1 (hGST A1-1) were made and characterized to determine the structural determinants for alkenal activity. The choice of mutations was based on primary structure alignments of hGST A1-1 and the Alpha class enzyme with the highest alkenal activity, hGST A4-4, from three different species and crystal structure comparisons between the human enzymes.
