  1. 1. In Vitro Studies of Adrenocorticolytic DDT Metabolites, with Special Focus on 3-methylsulfonyl-DDE

    University dissertation from Uppsala, Sweden : Acta Universitatis Upsaliensis

    Author : Vendela Asp; Uppsala universitet.; [2010]
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Toxicity; 3-methylsulfonyl-DDE; CYP; adrenal cortex; metabolic activation; steroid hormones; Y-1; H295R; mitotane; adrenocortical carcinoma; NATURAL SCIENCES Biology Cell and molecular biology Toxicology; NATURVETENSKAP Biologi Cell- och molekylärbiologi Toxikologi; Biologi med inriktning mot ekotoxikologi; Biology with specialization in Environmental Toxicology;

    Abstract : The DDT metabolite 3-methylsulfonyl-DDE (3-MeSO2-DDE) is bioactivated by cytochrome P450 11B1 (CYP11B1) in the adrenal cortex of mice and forms irreversibly bound protein adducts, reduces glucocorticoid secretion, and induces cell death selectively in cortisol-producing adrenocortical cells. 3-MeSO2-DDE has therefore been proposed as a lead compound for an improved adrenocortical carcinoma (ACC) therapy. 2. 2. Retinoic Acid Metabolism Blocking Agents and the Skin In vivo and in vitro Studies of the Effects on Normal and Diseased Human Epidermis

    University dissertation from Uppsala : Universitetsbiblioteket

    Author : Elizabeth Pavez Loriè; Uppsala universitet.; [2008]
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; CYP; CYP26; retinoids; vitamin A; RAMBA; metabolism; keratinocyte; epidermis; retinoid regulated genes; MEDICINE Dermatology and venerology; clinical genetics; internal medicine Dermatology and venerology; MEDICIN Dermatologi och venerologi; klinisk genetik; invärtesmedicin Dermatologi och venerologi;

    Abstract : Retinoic Acid Metabolism Blocking Agents (RAMBAs) increase the endogenous levels of all-trans retinoic acid (RA) by inhibiting CYP26 enzymes. Thus they are believed to mimic the effects of retinoid treatment. Their mechanism of action and effects on vitamin A metabolism in keratinocytes are however uncertain. READ MORE

  3. 3. Steroid-Metabolizing Cytochrome P450 (CYP) Enzymes in the Maintenance of Cholesterol and Sex Hormone Levels

    University dissertation from Uppsala : Acta Universitatis Upsaliensis

    Author : Hanna Pettersson; Uppsala universitet.; [2009]
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; CYP27A1; CYP7B1; cytochrome P450; estrogen receptor; androgen receptor; cholesterol; sex hormone; hydroxylation; steroid metabolism; regulation of steroid levels; PHARMACY Pharmaceutical biochemistry; FARMACI Farmaceutisk biokemi; Farmaceutisk biokemi; Pharmaceutical Biochemistry;

    Abstract : The enzymes CYP27A1 and CYP7B1 are widely expressed in various human tissues and perform catalytic reactions in cholesterol homeostasis and endocrine signaling. We have investigated the metabolism of a synthetic oxysterol. READ MORE

  4. 4. Novel Fatty Acid Dioxygenases of Human and Plant Pathogenic Fungi Studies by Gene Deletion and Expression

    University dissertation from Uppsala : Acta Universitatis Upsaliensis

    Author : Fredrik Jernerén; Uppsala universitet.; [2011]
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; aspergilli; dioxygenase; oxygenase; Magnaporthe oryzae; Gaeumannomyces graminis; Lasiodiplodia theobromae; jasmonic acid; linoleate diol synthase; cyclooxygenase; prostaglandin H synthase; cytochrome P450; oxylipin; hydroperoxide isomerase; allene oxide synthase; PHARMACY Pharmaceutical pharmacology; FARMACI Farmaceutisk farmakologi; Biochemical Pharmacology; Biokemisk farmakologi;

    Abstract : The dioxygenase-cytochrome P450 fusion proteins (DOX-CYP) comprise a heme-containing enzyme family that shares structural and catalytic properties with mammalian prostaglandin H (PGH) synthases. 7,8-Linoleate diol synthase (7,8-LDS) of Gaeumannomyces graminis was first characterized, and DOX-CYP enzymes are of mechanistic and biological interest.
