Abstract : Cytochrome P450 (CYP) enzymes in extrahepatic tissues may play important roles in regulating the capacity of individual cells to metabolize environmental carcinogens and hormones. In this thesis, we characterized the cytochrome P450 profile of the human normal breast and investigated whether polymorphisms in the genes encoding the estrogen metabolizing enzymes CYP1131 and Catechol-O-methyltransferase (COMT) were associated with breast or endometrial cancer risk. Abstract : Cyclophosphamide (CPA) is widely used anti-cancer drug that has to be catalyzed by the cytochrome P450 (CYP) enzyme 2B6 as well as some other CYPs to exert its cytotoxic activity. A high degree of interindividual variation in CPA kinetics has been reported in both children and adult patients, which may be due to several factors such as concomitantly used drugs, variation in physiological status of individual patients and genetic polymorphism of the CYP enzymes.
