  1. 6. Pharmacokinetic consequences of CYP2D6 genotypes with emphasis on gene duplication / amplification

    Author : Per Dalén; Karolinska Institutet; Karolinska Institutet; []
    Keywords : Drug metabolism; CYP2D6. debrisoquine; nortriptyline; quinidine; genotype; phenotype; cytochrome P450; gene duplication; gene amplification;

    Abstract : The relationship between the major Caucasian and Oriental CYP2D6 genotypes and the disposition of two CYP2D6 substrates nortriptyline and debrisoquine was investigated. Caucasians with 0, 1, 2, 3/4 and 13 functional CYP2D6-genes as well as Chinese and Korean subjects homo- and heterozygous for the CYP2D6*1 and CYP2D6*10 alleles were included. READ MORE

  2. 7. CYP2D6-polymorphism and effect of adjuvant tamoxifen in breast cancer patients

    Author : Linda Thorén; Karolinska Institutet; Karolinska Institutet; []
    Keywords : ;

    Abstract : Adjuvant tamoxifen at the standard dose of 20 mg daily for five to ten years reduces the risk for relapse and mortality in hormone sensitive breast cancer. The effect however varies and no early marker of poor response is yet available. READ MORE

  3. 8. Drug metabolic capacity in Koreans : CYP2D6 & CYP2C19 pheno- and genotype relationships in healthy volunteers and in patients

    Author : Hyung-Keun Roh; Karolinska Institutet; Karolinska Institutet; []
    Keywords : Drug metabolism; phenotype; genotype; CYP2D6; CYP2C 19; Korean; haloperidol; risperidone; omeprazole; gastric pH; gastrin; schizophrenia; patient.;

    Abstract : This thesis aimed to investigate the relationship between pheno- and genotype of two drugmetabolizing enzymes CYP2D6 and CYYP2C19 in Koreans. Population studies were conducted in healthy volunteers to obtain basic information on these two enzymes. READ MORE

  4. 9. Enantioselective drug metabolism catalysed by the polymorphic CYP2D6 and CYP2C19 : a methodological investigation focused on mianserin, mephenytoin and omeprazole

    Author : Gunnel Tybring; Karolinska Institutet; Karolinska Institutet; []
    Keywords : Drug metabolism; CYP2D6; CYP2CI9; mianserin; mephenytoin; omeprazole; enantiomers; chiral analysis;

    Abstract : The aim of this thesis was to develop enantioselective analytical methods and apply them to studies of oxidative metabolism of chiral drugs catalysed by the polymorphic cytochrome P450 CYP2D6 and CYP2C 19 enzymes. The work has been focused on mianserin, mephenytoin and omeprazole. READ MORE

  5. 10. Experimental and computational investigation of affinity and selectivity factors in CYP2D6 and CYP3A4 mediated metabolism

    Author : Britta Bonn; Göteborgs universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Cytochrome P450; in vitro metabolism; biotransformation; computational modeling; drug design;

    Abstract : The assessment of ADME properties and metabolic behavior of a drug is central in drug discovery and drug design. The main target for studies of metabolic properties is the Cytochrome P450 (CYP), which is responsible for the metabolism of a majority of drugs on the market and consequently also involved in many drug-drug interactions.
