  1. 1. Studies on the mechanism of regulation of bile acid synthesis in humans with some aspects on genetic factors

    Author : Anna Abrahamsson; Karolinska Institutet; Karolinska Institutet; []
    Keywords : Bile acid synthesis; CYP7A1; CYP8B1; nuclear factors; primary human hepatocytes; regulation; polymorphism;

    Abstract : In the present investigation we studied human bile acid synthesis and its regulation both in vivo and in vitro and also evaluated a possible influence of polymorphisms in the CYP7A1 and CYP8B1 genes on bile acid synthesis in vivo. The following observations were made: · Primary human hepatocytes cultured on matrigel under serum-free conditions produced bile acids consistent with the pattern in vivo, i. READ MORE

  2. 2. Studies on the transcriptional regulation of sterol 12alpha-hydroxylase (CYP8B1)

    Author : Yizeng Yang; Karolinska Institutet; Karolinska Institutet; []
    Keywords : Bile acids; CYP8B1; CYP7A1; transcription; nuclear receptor; PPAR-alpha; HNF4-alpha; FTF; SREBP; thyroid hormones.;

    Abstract : The conversion of cholesterol into bile acids is mainly via two pathways, which are initiated by CYP7A1 and CYP27A1 respectedly. CYP8B1, a microsomal P450 cytochrome, hydroxylates of the steroid nucleus at the C-12alpha position to form 7alpha, 12alpha-dihydroxy-4-cholesten-3-one, a facultative precursor of CA. READ MORE

  3. 3. Studies on cholesterol homeostasis by modulation of lipoproteins and bile acids

    Author : Lisa-Mari Nilsson; Karolinska Institutet; Karolinska Institutet; []
    Keywords : Cholesterol; lipoproteins; bile acids; cholestyramine; chenodeoxycholic acid; CYP7A1; HNF4alpha; PCSK9; hepatocytes; ethanol; atherosclerosis; ApoE; thyroid hormone receptor;

    Abstract : Excess plasma cholesterol, in particular high levels of LDL cholesterol, is a major risk factor for the development of atherosclerosis. The liver has a central role in the regulation of cholesterol metabolism: i) a major part of the cholesterol present in the body is synthesized in the liver; ii) the level of plasma lipoproteins is regulated by liver uptake and secretion of these particles and iii) the liver is essential for elimination of cholesterol from the body as such or by its conversion into bile acids. 4. 4. Studies on lipoprotein metabolism and atherogenesis

    Author : Katharina Slätis; Karolinska Institutet; Karolinska Institutet; []
    Keywords : ApoA1; ApoB; ApoE; atherosclerosis; bile acids; cholic acid; cholesterol; cholesterol absorption; CYP7A1; CYP8B1; dexamethasone; GC-1; PAI-1; thyroid hormone receptor; very low density lipoprotein receptor;

    Abstract : Atherosclerosis is the primary cause of cardiovascular events such as angina pectoris and myocardial infarction, which together with stroke are responsible for around 50% of all deaths in the United States and Europe. Usually atherosclerosis develops over several decades and leads to a progressive narrowing of arterial vessels. READ MORE

  5. 5. Studies on the hormonal regulation of bile acid synthesis

    Author : Thomas Lundåsen; Karolinska Institutet; Karolinska Institutet; []
    Keywords : ;

    Abstract : The maintenance of a normal turnover of cholesterol is of vital importance, and disturbances of cholesterol metabolism may result in several important disease conditions. The major route for the elimination of cholesterol from the body is by hepatic secretion of cholesterol and bile acids (BAs) into the bile for subsequent fecal excretion. READ MORE
