  1. 6. The Role of Phosphodiesterase 3B in the Regulation of Insulin Secretion

    Author : Linda Härndahl; Institutionen för experimentell medicinsk vetenskap; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; diabetology; Endokrinologi; sekretion; diabetologi; secreting systems; Endocrinology; beta-cell; diabetes; islet; PDE3B; cAMP;

    Abstract : Pancreatic beta-cell dysfunction and insulin resistance are the two hallmarks of type 2 diabetes. An early sign of beta-cell dysfunction is impaired nutrient-induced insulin release. Several insulin secretagogues act by increasing the formation of intracellular cAMP. READ MORE

  2. 7. Cyclic nucleotide signalling systems in vascular smooth muscle cells and immune system cells with special reference to phosphodiesterases PDE3 and PDE4

    Author : Dag Ekholm; Institutionen för experimentell medicinsk vetenskap; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; HTLV-I; multiple sclerosis; proliferation; phosphorylation; T-lymphocytes; vascular smooth muscle; cloning; protein kinase A; cGMP; cAMP; PDE4; Phosphodiesterase; PDE3; cilostamide; rolipram; Histology; cytochemistry; histochemistry; tissue culture; Histologi; cytokemi; histokemi; vävnadskultur;

    Abstract : The cyclic nucleotide signalling systems are important in vascular smooth muscle and in the immune system, and are of interest from a pharmacological point of view. Drugs which interfere with the cyclic nucleotide levels have profound effects on heart, blood vessels and airways and have actual or potential use in several important diseases, such as heart failure, hypertension, arteriosclerosis, asthma and autoimmune diseases. 3. 8. Epac2 signaling at the β-cell plasma membrane

    Author : Ida Alenkvist; Anders Tengholm; Olga Göransson; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; β-cell; insulin secretion; cAMP; Ca2 ; Epac; Rap; exocytosis; sulfonylurea; Medical Cell Biology; Medicinsk cellbiologi;

    Abstract : Secretion of appropriate amounts of insulin from pancreatic β-cells is crucial for glucose homeostasis. The β-cells release insulin in response to glucose and other nutrients, hormones and neurotransmitters, which trigger intracellular signaling cascades, that result in exocytotic fusion of insulin-containing vesicles with the plasma membrane. 4. 9. α-Cell signalling in glucose-regulated glucagon secretion

    Author : Qian Yu; Anders Tengholm; Erik Gylfe; Sebastian Barg; David W Piston; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Islets; glucagon; insulin; somatostatin; ATP; cAMP; Ca2 ; GHB; oscillations; α-cell; β-cell; δ-cell; Medical Cell Biology; Medicinsk cellbiologi;

    Abstract : Glucagon is a blood glucose-elevating hormone released from α-cells in the islets of Langerhans during hypoglycaemia. Glucagon is critical for glucose homeostasis and inappropriate regulation of its secretion underlies both impaired counter-regulation of hypoglycaemia and chronic hyperglycaemia in diabetes patients. 5. 10. Cyclic Nucleotide Phosphodiesterase 3B: Regulation in rat adipocytes and 3T3-L1 cells

    Author : Tova Landström; Institutionen för experimentell medicinsk vetenskap; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; 3T3-L1 TNF-alpha; adipocyte; insulin; p70S6 kinase; mitogen-activated protein kinase; protein kinase B; phosphatidylinositol-3 kinase; phosphodiesterase 3B; cAMP; lipolysis; phosphorylation; signal transduction; Biochemistry; Metabolism; Biokemi; metabolism;

    Abstract : Insulin stimulation of rat adipocytes results in phosphorylation and activation of the cyclic nucleotide phosphodiesterase 3B (PDE3B), a key enzyme in the antilipolytic signalling pathway of this hormone. In this thesis, the site phosphorylated in PDE3B upon stimulation of rat adipocytes with insulin and/or the beta-adrenergic agonist, isoproterenol is identified. READ MORE
