  1. 1. Chemical attenuation of bacterial virulence : small molecule inhibitors of type III secretion

    Author : Anna Kauppi; Mikael Elofsson; Anders Karlén; Umeå universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; antibiotic resistance; virulence; type III secretion; T3SS inhibitors; Yop; SMD; QSAR; screening; Yersinia pseudotuberculosis; Organic chemistry; Organisk kemi;

    Abstract : Despite the large arsenal of antibiotics available on the market, treatment of bacterial infections becomes more challenging in view of the fact that microbes develop resistance against existing drugs. There is an obvious need for novel drugs acting on both old and new targets in bacteria. READ MORE

    Author : Daniel Decker; Leszek A. Kleczkowski; Henrik Vibe Schelle; Umeå universitet; []
    Keywords : LANTBRUKSVETENSKAPER; AGRICULTURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; Chemical library screening; Cell wall synthesis; Glycosylation; Nucleotide sugars; Oligomerization; Protein structure; Reverse chemical genetics; Sugar activation; UDP-sugar synthesis; biokemisk farmakologi; biochemical pharmacology; biokemi; Biochemistry; biology; biologi; molekylärbiologi; Molecular Biology;

    Abstract : UDP-sugar metabolizing pyrophosphorylases provide the primary mechanism for de novo synthesis of UDP-sugars, which can then be used for myriads of glycosyltranferase reactions, producing cell wall carbohydrates, sucrose, glycoproteins and glycolipids, as well as many other glycosylated compounds. The pyrophosphorylases can be divided into three families: UDP-Glc pyrophosphorylase (UGPase), UDP-sugar pyrophosphorylase (USPase) and UDP-N-acety lglucosamine pyrophosphorylase (UAGPase), which can be discriminated both by differences in accepted substrate range and amino acid sequences. 3. 3. Simplified Routines for Sample Preparation and Analysis of Chemical Warfare Agent Degradation Products

    Author : Raja Subramaniam; Calle Nilsson; Paula Vanninen; Umeå universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Nerve agents; Alkyl alkylphosphonic acids; Solid phase derivatization; Fluorinated diazomethane reagent; High-energy collision induced dissociation; CID MS MS library; analytisk kemi; Analytical Chemistry;

    Abstract : The thesis describes the development of new and improved methods for analyzing degradation markers from organophosphorus Chemical Warfare Agents (CWAs). Paper I and II describes an innovative and significantly improved method for the enrichment, derivatization (trimethysilylation) and GC-MS analysis of a broad range of organophosphorus CWAs degradation markers, namely the alkylphosphonic acids and a zwitterionic compound. Abstract : Antibiotic resistance has evolved significantly to become one of the serious threats to public health today. Yet, the pipeline of new antibiotics is drying up and is lagging behind the challenging needs. READ MORE

  5. 5. A Novel Route for Construction of Multipurpose Receptors through Chemical Modification of Glutathione Transferases

    Author : Johan Viljanen; Bengt Harald Jonsson; Kerstin Broo; Sophia Hober; Linköpings universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Human GST A1-1; site-specific covalent modification; tyrosine 9; lysine 216; methionine 208; multipurpose receptor; pattern recognition; protein purification; Chemistry; Kemi;

    Abstract : This thesis describes how the human Alpha class glutathione transferase (GST) A1-1 can be reprogrammed either to function as a multipurpose biosensor for detection of small molecule analytes, or as a handle providing for more efficient protein purification.A novel, user-friendly, and efficient method for site-specific introduction of functional groups into the active site of hGST A1-1 is the platform for these achievements.
